Objective:(1)to detect the difference of serum metabonomics among patients with pancreatic cancer,chronic pancreatitis and healthy people by using gas chromatography-mass spectrometry(GC-MS).(2)using gas chromatography-mass spectrometry(GC-MS)metabonomics to provide a new method for distinguishing chronic pancreatitis from healthy controls.Further analysis of differential metabolites can provide reference significance for further research on the occurrence and development of pancreatic cancer.Methods:(1)the sera of 37 patients with pancreatic cancer,33 patients with chronic pancreatitis and 32 healthy subjects were collected from Nanfang Hospital of Southern Medical University from May 2020 to August 2020.Patients with pancreatic cancer were treated as pancreatic cancer group(pancreaticcancer,PC),chronic pancreatitis as benign pancreatic disease group(chronicpancreatitis,PT),and healthy subjects as control group(Normalcontrol,NC).(2)the sera of pancreatic cancer group,chronic pancreatitis group and healthy control group were analyzed by gas chromatography-mass spectrometry(GC-MS).Principal component analysis(Principalcomponentanalysis,PCA)and OPLS-DA were used to screen the differential serum metabolic markers.Further analyze the correlation between differential metabolites and the correlation with patient test results,and compare the relative abundance of differential metabolites in each group,and finally use the random forest classification model to further analyze the contribution ranking of metabolites.Results:(1)A total of 43 differential metabolites were screened between patients with pancreatic cancer,chronic pancreatitis and healthy controls.They are lactic acid,valine,valine 3-hydroxypropionic acid,methanol phosphate,2-hydroxyvaleric acid,phosphate,succinic acid,veratrol acid,alanine,tartaric acid,serine,L-Besthreonine,oxyproline,salicylic acid,adipic acid,creatine 4-aminophenol,lauric acid,L-glucan,epoxy resin,galactoic acid,methyl linoleate,methyl stearate,indole-3-lactic acid,tryptophan,stearic acid.Linoleic acid,elemic acid,stearic acid,arachidonic acid.(2)the correlation between differential metabolites is different.In addition,the differential metabolites positively correlated with CA199 include tryptophan,serine,ornithine,alanine,3-phosphoglyceric acid and so on.(3)Veratrinic acid,gluconolactone,creatine and fructose gradually decreased in healthy people,chronic pancreatitis and pancreatic cancer,and the relative abundance in healthy people was much higher than that in chronic pancreatitis and pancreatic cancer.Oleinic acid,lactic acid,ribose alcohol,lauric acid,terephthalic acid,fatty acid and myristic acid also showed a decreasing trend in healthy people,chronic pancreatitis and pancreatic cancer groups.And it showed a low relative abundance in the pancreatic cancer group.The abundance of L-glucan is the highest in pancreatic cancer,chronic pancreatitis and healthy people.Serine,hexadecanoic acid,linoleic acid methyl,ornithine and α-glucosamine have relatively high abundance in chronic pancreatitis and pancreatic cancer.Fructose,stearic acid,lactic acid,lauric acid,stearic acid,diglycerol,arabinol,arachidonic acid,linoleic acid,valine,tartaric acid,phosphate and tyrosine have relatively low abundance in pancreatic cancer.(4)the average accuracy of terephthalic acid,L-glucan,fatty acid,methanol phosphate,hexadecanoic acid,octadecanoic acid,linoleic acid,tryptophan,arachidonic acid,L-trimethylphenylalanine,stearic acid,glycerol 3-phosphate,gluconic acid,alanine and ornithine decreased.Conclusion:there is a significant difference in serum metabonomics among pancreatic cancer,chronic pancreatitis and healthy groups,and the difference of metabolites suggests that the amino acid,energy and lipid metabolism of pancreatic cancer have changed.metabolic pathway-induced metabolic abnormalities have a certain reference value for candidate markers for the diagnosis of pancreatic cancer. |
