Objective:(1)To analyze the expression of long chain non-coding RNACCAT1(Lnc-CCAT1)and sex-determining region Y-box protein 2(SOX2)in gastric cancer,and to study the relationship between long-chain non-coding protein(Lnc-CCAT1)and clinicopathological features of gastric cancer and its effect on prognosis.(2)To clarify the relationship between Lnc-CCAT1 and SOX2 and their regulation mechanism on the invasion and metastasis of gastric cancer cells.Methods:(1)50 cases of gastric cancer and corresponding paracancerous tissues were detected.The expression levels of Lnc-CCAT1 and SOX2mRNA were determined by real-time fluorescence quantitative PCR(qPCR),and the expression of SOX2 protein was detected by Western blotting(WB).(2)The expression of Lnc-CCAT1 in gastric cancer cell lines HGC-27,SGC-790 and MKN-45 was detected by qPCR.(3)The relationship between the expression of LncCCAT1 and SOX2 and the clinicopathological characteristics of gastric cancer patients was analyzed by collecting clinical data.The factors affecting the expression of Lnc-CCAT1 in gastric cancer were analyzed by univariate and multivariate logistic regression analysis.The relationship between the expression of LncCCAT1,SOX2 and overall survival rate(OS)was analyzed by online database Kaplan-Meier(KMMI)Plotter.(4)The effect of quantitative Lnc-CCAT1 on cell cycle and apoptotic behavior of gastric cancer cells was detected by flow cytometry.(5)Transwell invasion test and scratch healing test were used to detect the effect of Lnc-CCAT1 on the migration and invasive behavior of gastric cancer cells.(6)To silence the expression of LncCCAT1 in gastric cancer cell line HGC-27,to detect its effect on the expression of SOX2,and to explore the functional relationship between CCAT1 and SOX2.Results:(1)The expression level of Lnc-CCAT1 in gastric cancer tissues was significantly higher than that in paracancerous tissues(P<0 05).There was a close relationship between Lnc-CCAT1 expression and tumor diameter,TNM stage,lymph node(LN)metastasis and depth of invasion.The expression of SOX2 in gastric cancer was significantly lower than that in paracancerous tissues,and it was significantly correlated with tumor diameter,depth of invasion,degree of differentiation,lymph node metastasis and TNM stage.Kmurm survival analysis showed that patients with high Lnc-CCAT1 expression had shorter survival time and worse prognosis.(2)the expression of Lnc-CCAT1 in gastric cancer cell line HGC-27 was higher than that in other cells.(3)Compared with the paracancerous tissues,the expression of SOX2mRNA in gastric cancer was lower,and it was negatively correlated with the expression of Lnc-CCAT1.(4)Silencing the expression of Lnc-CCAT1 can promote apoptosis,and silencing the expression of CCAT1 will inhibit the invasion and migration of gastric cancer cells.(5)Lnc-CCAT1 may be involved in the regulation of SOX2 protein expression activity.Conclusion:(1)The high expression of Lnc-CCAT1 in gastric cancer is related to TNM stage,tumor diameter,depth of invasion and LN metastasis.Tumor size can be regarded as an independent risk factor for the expression of Lnc-CCAT1 in gastric cancer.The low expression of SOX2 in gastric cancer is related to TNM stage,differentiation,tumor diameter,LN metastasis and depth of invasion,and the degree of differentiation can be regarded as an independent risk factor for SOX2 expression in gastric cancer.(2)The overall survival rate of patients with high expression of Lnc-CCAT1 was significantly lower than that of patients with low expression of SOX2.Although low expression of SOX2 was found in gastric cancer,the prognosis of patients with high expression of VEGF was worse.(3)Lnc-CCAT1 may affect the biological behavior of gastric cancer cells by regulating the expression of SOX2 protein.These results contribute to a better understanding of the mechanism of Lnc-CCAT1/SOX2 pathway in order to find new potential therapeutic targets.
