Study On The Effect Of Platelet Rich Plasma (PRP) On Preventing Skin Photoaging In Rats

Objective:After the rats were pretreated with platelet rich plasma(PRP),the changes of ROS activity,MDA,CTA,SOD,GSH Px activity,collagen I,MMP-1,TGF-β1 in the skin of rats were observed by UV induction.Methods:Forty healthy female rats were randomLy divided into four groups with 10 rats in each group.Group A:blank control group(normal control group);Group B:UV irradiation without drying;Group C:UV irradiation+saline pre-treatment group(20 points are distributed in the depilated area of the back of rats on average,and 0.1 mL of saline is injected at each point),once every four weeks,twice in total;Group D:UV irradiation+PRP pre-treatment group(the average distribution of the hair removal area on the back of rats is 20 points,and each point is injected with 0.1 mL of PRP),once every four weeks,twice in total.C and D group was treated with normal saline and PRP respectively and then treated with ultraviolet radiation for 8 weeks with group B.After 8 weeks,the back skin of rats in each group was dynamically observed with naked eyes;The content of ROS in rat skin was detected by flow cytometry;The contents of CTA,SOD,GSH-Px and MDA in rat skin were measured by Elisa method;Western blot analysis of the effect of PRP pretreatment on MMP-1 and TGF-β1;The expression of collagen I was detected by immunohistochemistry.Result:(1)After 8 weeks of UV irradiation,compared with group A,the skin of group B had poor elasticity,roughness,dry scaling,wrinkle formation and pigmentation.Compared with group B and group C,group D had good elasticity,moist and smooth skin appearance,no pigmentation,no skin swelling,no wrinkles,no flushing.There was no significant difference in skin appearance between group B and group C.(2)Flow cytometry showed that the fluorescence intensity of group B was significantly higher than that of group A(P<0.01);B.Group C was significantly higher than group D(P<0.01);Group B was higher than group C(P>0.05),and the difference was not statistically significant.(3)Elisa method showed that the activities of CTA,SOD and GSH Px in group B were lower than those in group A(P<0.01),and the difference was statistically significant;The activities of CTA,SOD and GSH Px in group D were significantly higher than those in groups B and C(P<0.01);There was no significant difference between group B and group C(P>0.05).The content of MDA in group B was significantly higher than that in group A(P<0.05);Group D was lower than group B and C(P<0.05),the difference was statistically significant;There was no significant difference between group B and group C(P>0.05).(4)Western blot showed that the expression of MMP-1 in group B was significantly higher than that in group A(P<0.01);The expression of MMP-1 in group D was significantly lower than that in group B and C(P<0.01);There was no significant difference between group B and group C(P>0.05).TGF-β The expression of 1 in group B was significantly lower than that in group A(P<0.01);TGF in group D compared with group B and C-β The expression of 1 was significantly increased(P<0.01);(5)The results of immunohistochemistry showed that the average optical density of type Ⅰ collagen in group B was significantly lower than that in group A(P<0.05);The average optical density of type Ⅰ collagen in group D was significantly higher than that in group B and C(P<0.05);There was no significant difference between group B and group C(P>0.05).Conclusion:PRP pretreatment can inhibit the production of ROS and MDA in the skin after UV irradiation,increase the activities of antioxidant enzymes SOD,CAT and GSH Px,and inhibit oxidative stress reaction.PRP pretreatment can down regulate the expression of MMP-1 and up regulate TGF-β1,promote the synthesis of collagen,prevent or delay the occurrence of light aging.
