Objective:To investigate the effect of benazepril on the proliferation and differentiation of urine-derived stem cells(USC)in children with chronic kidney disease(CKD).Methods:CKD caused by nephrotic syndrome was used as the representative disease of CKD in children.Urine was collected from normal children and children with CKD,and USC were isolated,cultured,and identified from urine sources.The proliferation,differentiation ability,angiotensin converting enzyme(ACE),inflammatory factors(IL-1β,connexin43)and reactive oxygen species(ROS)levels in oxidative stress were compared between the two groups of USC.Different concentrations of benazepril were given to interfere with USC,and the appropriate concentration was screened.The effect of Benazepril on the proliferation and differentiation of USC in children with chronic kidney disease(CKD)was examined after intervention at appropriate concentration.Results:1.USC can be grown from urine of both normal and CKD children.However,USC from CKD children can only be passaged to the fifth generation,and their ability of proliferation and differentiation into renal tubular epithelial cells is weaker than that of normal children.2.ACE is detected in USC of both normal children and children with CKD,and the level is higher in USC of children with CKD.3.The levels of inflammatory factors IL-1β,Connexin 43 and oxidative stress product ROS in USC of children with CKD are higher than those of normal children.4.1 μmol/L benazepril can promote the proliferation of USC in children with CKD.Although other concentrations of benazepril can promote the proliferation of USC in the early stage,these effects are gradually lost or even become inhibitory with the prolongation of administration time.5.After treatment with 1 μmol/L benazepril,the differentiation ability of USC from CKD children into renal tubular epithelial cells was increased,and the levels of ACE,inflammatory factors IL-1β,Connexin 43 and oxidative stress product ROS were decreased.Conclusion:The levels of ACE,IL-1β,Connexin 43 and oxidative stress product ROS are increased in USC from CKD children,which may be important reasons for the decreased ability of proliferation and differentiation into renal tubular epithelial cells of USC.1 μmol/L Benazepril could reduce the levels of ACE,inflammatory response and oxidative stress,and improve the impaired proliferation and differentiation ability.
