Objective The aim of this study was to correlate the expression of ATP-binding cassette transporter subfamily A member 8(ABCA8)with the clinicopathological parameters of colorectal cancer(CRC),to investigate the relationship between ABCA8 expression and colorectal cancer metastasis and prognosis,and to provide new ideas for the prevention,diagnosis and targeted treatment of colorectal cancer.Methods In this study,the expression of ABCA8 mRNA in CRC tissues and normal paired tissues was detected by fluorescent quantitative PCR(RT-PCR)method.Immunohistochemistry(IHC)staining was used to detect the expression of ABCA8 protein in CRC tissues,and the expression was statistically analyzed with clinicopathological parameters(age,gender,tumor size,tumor primary site,lymph node metastasis,degree of histological differentiation and general types).Kaplan-Meier survival analysis and multifactorial COX proportional risk model were applied to analyze the relationship between ABCA8 expression and CRC prognosis.A lentiviral vector was used to construct a stable ABCA8 overexpressing colorectal cancer RKO cell line,and the effect of ABCA8 on colorectal cancer cell migration was investigated by transwell assay and scratch healing assay.Results The mRNA expression level of ABCA8 was significantly lower in colorectal cancer tissues than in paired normal tissues(P<0.05).The protein expression of ABCA8 in colorectal cancer tissues was negatively correlated with lymph node metastasis,and the difference was statistically significant(P<0.05);ABCA8 was not significantly correlated with age,gender,tumor size,tumor primary site,degree of histological differentiation and general types,and the difference was not statistically significant(P>0.05).the results of Kaplan-Meier survival analysis showed that The overall survival rate of patients with high ABCA8 expression was significantly higher than that of patients with low ABCA8 expression(P<0.05),and the results of the multifactorial COX proportional risk model analysis showed that ABCA8 was an independent protective factor for the prognosis of CRC patients.The results of the Transwell assay showed that the number of RKO cell migration after overexpression of ABCA8 was significantly lower than that of the control group(P<0.05),and the scratch The results of the healing assay showed that the area of scratch healing between RKO cells was significantly lower than that of the control group after overexpression of ABCA8(P<0.05).Conclusion ABCA8 expression was downregulated in CRC tissues,patients with low ABCA8 expression were more likely to develop lymph node metastasis,and low ABCA8 expression was associated with poor patient prognosis.The results of in vitro cytological experiments showed that ABCA8 was able to inhibit the migration of CRC cells.
