Objective:In this study,umbilical moxibustion was used to intervene patients with subthreshold depression in order to observe the clinical effect of umbilical moxibustion on subthreshold depression and analyze the correlation and difference of intestinal flora between patients with subthreshold depression and healthy people.To systematically study the effect of umbilical moxibustion on intestinal flora in patients with subthreshold depression.Methods:1.During the period from July 2022 to December 2022,the patients with subthreshold depression and healthy people were recruited from the Preventive Treatment Center of the Affiliated Hospital of Shandong University of Traditional Chinese Medicine as research subjects,and were divided into subthreshold depression group(AQ group)(n=36 cases)and healthy control group(HC group)(n=36 cases).Both groups of subjects were required to collect stool samples and conduct Center for Epidemiological Survey,Depression Scale(CES-D)and Hamilton Depression Scale 17-item(HAMD-17)scores.The samples were sequenced with 16S rDNA by Majorbio Bio-Pharm Technology Co.Ltd.(Shanghai,China).Through bioinformatics analysis,the similarities and differences of the structure of intestinal flora between AQ group and HC group were compared.2.AQ group was scored with CES-D and HAMD-17 after 8 times of navel moxibustion treatment(once a week),and stool samples were collected,and 16S rDNA sequencing was completed by Majorbio Bio-Pharm Technology Co.Ltd.(Shanghai,China).SPSS 26.0 statistical software was used for data analysis to evaluate the clinical effect of umbilicus moxibustion on subthreshold depression.The sequencing results were analyzed by bioinformatics to explore the effect of navel moxibustion on the intestinal flora of patients with subthreshold depression.Results:1.Comparison between subliminal depression group before umbilical moxibustion(AQ group)and healthy control group(HC group):The scores of CES-D and HAMD-17 in AQ group were significantly higher than those in HC group(P<0.05).The sobs index,shannon index and ace index of AQ group were significantly lower than those of HC group(P<0.05),while simpson index was higher.The dominant species were similar in the community composition at the phylum level and the genus level,but the abundance of each species was different.PLS-DA analysis showed that the diversity of intestinal flora in AQ group and HC group samples was significantly different at the genus level.LEfSe multi-level species difference discriminant analysis showed that the intestinal flora with significant influence in AQ group mainly belonged to Proteobacteria,and the intestinal flora with significant influence in HC group mainly belonged to Fir=micutes.The abundance of Escherichia-Shigella in AQ group was higher than that of HC group(P<0.05).The abundance of Eubacterium-hallii_group,Ruminococcus,Christensenellaceae_R-7_group,Paraprevotella was lower than that of HC group(P<0.05).2.Comparison between the post-umbilical-moxibustion(AH group)and pre-umbilical-moxibustion(AQ group)of the subthreshold depression group:The CES-D and HAMD-17 scores of AH group were lower than AQ group,the difference was statistically significant(P<0.05).The effective rate of navel moxibustion in treating subthreshold depression was 90.91%.The sobs index,shannon index,ace index and chao index of AH group were higher than AQ group.The dominant species were roughly similar in the community composition at the gate level and the genus level,and the abundance of each species was different.PLS-DA analysis showed that at the genus level,more sample points of AH group had obvious displacement to HC group.The abundance of Escherichia-Shigella in AH group was lower than that of AQ group(P<0.05),and the abundance of Ruminococcus,Christensenellaceae_R-7_group,Paraprevotella in AH group was higher than that of AQ group(P<0.05).Conclusion:The patients with subthreshold depression have appeared intestinal flora disorder,which is manifested by the decrease of the abundance and diversity of the flora,although it is similar to the dominant species in healthy people,but the abundance of harmful bacteria increases and the abundance of beneficial bacteria decreases.Umbilical moxibustion can not only effectively improve the symptoms of patients with subthreshold depression,but also increase the abundance of beneficial bacteria and reduce the abundance of harmful bacteria in the intestinal flora of patients with subthreshold depression.
