| The scientific analysis of the processing principle of Mongolian medicine and the correlation study of the processing process and the change of the mechanism of Mongolian medicine are one of the important contents of the modernization of Mongolian medicine.In order to provide experimental basis for standardized processing and quality standardization of Mongolian medicine veratrum,in this experiment,the chemical composition of Mongolian medicine veratrum was studied before and after processing,the processing principle was clarified,and the best processing technology of Mongolian medicine veratrum was established by optimizing the processing technology,at the same time,the quality control method of UPLC fingerprint of Mongolian medicine veratrum was established.This study has the following 3 contents:First,a UPLC method was established for the determination of veratrol in Mongolian medicine veratrol extract.Select Waters Symmetry C18(5 μ m 4.6 × 250mm)chromatographic column,with water(A)-acetonitrile(B)as the mobile phase for gradient elution.The gradient elution procedure is 0-3 min,5% B;3~30 min,40% B;30~35 min,95% B;35 to 50 min,95% B,flow rate 0.3 ml/min,column temperature 40 ℃,detection wavelength 320 nm.The regression equation for the peak area versus the concentration of veratrol in the control sample is y=184167.28x-35293840,r=0.9994.There is a good linear relationship between the concentration of veratrol in the range of 0.10 mg/ml to 0.50 mg/ml.According to the relevant provisions of the Chinese Pharmacopoeia(2020),a methodological study was conducted on precision,accuracy,repeatability,and stability.The RSD values of precision,accuracy,and stability are all less than 2.98%,indicating that this method meets the requirements for content determination.Firstly,the processing technology of Mongolian medicine Veratrum was studied.Using the content of veratrol as an indicator,two processing techniques(swill soaking and milk processing)of Mongolian medicine veratrol were optimized.The results showed that the optimal processing conditions for soaking Mongolian medicine veratrol swill were as follows: the processing temperature was 15 ℃,the solid-liquid ratio was 1:6,and the processing time was 6 hours.Under these conditions,the content of veratrol is 4.779 %.The optimal processing conditions for milk processing are as follows: processing temperature is 100 ℃,solid-liquid ratio is 1:10,and processing time is 10 minutes.Under these conditions,the content of veratrol is4.330 %.In the end,a UPLC fingerprint method for quality control of Mongolian medicine Veratrum officinalis was established.The established UPLC method was used to evaluate the similarity of 7 batches of Mongolian medicine samples of Veratrum officinalis in combination with the "Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System".There are 24 common peaks in the UPLC fingerprints of seven Mongolian medicinal herbs from different origins.The results of cluster analysis and principal component analysis showed that although the chemical components of Mongolian medicine Veratrum from different regions were consistent,their chemical component content varied significantly.This result may be related to factors such as the growth environment,collection season,and storage time of Mongolian medicine Veratrum. |
