Objective Mouse middle cerebral artery occlusion and reperfusion(MCAO/R)model was replicated,to study the effect and the biological mechanismof Huatan Tongluo Decoction in early intervention on cerebral ischemia-reperfusion injury.Methods 85 healthy male mouse were involved,aged 8-12 weeks with the body weight of 22-25 g.These mouse were randomly divided into the Sham group(Sham,consisted of 10 animals),and the Model group(Model),the Model with Huatan Tongluo Decoction high-dose group(HTTLD-H),the Model with Huatan Tongluo Decoction middle-dose group(HTTLD-M),the Model with Huatan Tongluo Decoction low-dose group(HTTLD-L)and the Model with Aspirin Enteric-coated Tablets group(AECT),Each group consisted of 15 animals.30 minutes after successful replication of MCAO/R Model,the groups were given intragastric administration,and the Sham group and Model group were given equal volume normal saline.After 7 days of intervention,the number of death,weight change and neurological deficit score of mice were counted;Neurological impairment score was used to evaluate the degree of neurological impairment in mice.The infarct volume of brain tissue was detected by TTC staining.The morphological changes of brain tissue were detected by HE staining.The expression levels of v WF,Fibrinogen,Claudin-5 and Occludin in brain tissue were detected by Western blot.The expression of v WF,GP Ⅸ,Fibrinogen,Occludin and Claudin-5 in brain tissue were detected by immunofluorescence method.Results 1.Cerebral ischemia reperfusion can lead to the death of mice.Huatan Tongluo Decoction can reduce the death rate of MCAO/R mice,especially in high and medium doses.2.The weight of mice after MCAO/R decreased to different degrees,and the difference was significant compared with Sham group(P < 0.01).Huatan Tongluo Decoction at medium and low doses could improve the degree of weight loss of mice,but the difference was not significant(P > 0.05).High dose administration caused a greater weight loss than Model group,and the difference was significant(P < 0.01).3.MCAO/R can cause neurological impairment in mice,Huatan Tongluo Decoction for 7 days can improve the neurological impairment in mice,especially in the medium dose group(P <0.01).4.Huatan Tongluo Decoction could reduce the infarct volume of brain tissue in MCAO/R mice,and the decrease was most significant in medium dose group(P < 0.01).5.Huatan Tongluo Decoction can maintain the integrity of blood vessel wall in the brain tissue of MCAO/R mice to a certain extent,improve blood circulation,and then maintain the survival of neurons,reduce death.6.MCAO/R resulted in increased expression of v WF(P < 0.01),excessive formation of platelet adhesion and aggregation(P < 0.01),Fibrinogen in mouse brain tissue(P < 0.01),and damage of blood-brain barrier,and decreased expression of Claudin-5 and Occludin in tissue(P < 0.05).Huatan Tongluo Decoction could reduce v WF expression in brain tissue and reduce platelet adhesion and aggregation.Reduce Fibrinogen deposition,maintain the expression of Claudin-5 and Occludin to a certain extent,and maintain the integrity of the blood-brain barrier.Conclusions Huatan Tongluo Decoction can reduce the mortality rate of cerebral ischemia reperfusion mice,reduce the infarct volume of brain tissue and improve nerve function,which may be related to Huatan Tongluo Decoction reducing the expression of v WF in brain tissue of cerebral ischemia reperfusion mice,reducing platelet adhesion and aggregation,and reducing Fibrinogen deposition.To some extent,Claudin-5 and Occludin expression were maintained,and the integrity of blood-brain barrier was maintained.
