ObjectiveMore and more studies have confirmed that microRNAs(miRNAs/miRs),as oncogenes or tumor suppressor genes,play an important regulatory role in the occurrence and development of various cancers.The purpose of this study was to investigate the potential roles and mechanisms of miR-124-3p and insulin like growth factor 2 mRNA binding protein(IGF2BP1)in the development of endometrial cancer.MethodsFirst,differential expression of miRNAs in ECs was identified from the GSE35794 and GSE25405 datasets using Venn diagrams.Next,a series of functional tests were performed to measure the ability of cancer cells to proliferate and migrate.Then,downstream target genes of miR-124-3p were identified using bioinformatics tools,and the predictions were validated by RT-qPCR,western blotting,and dual-luciferase reporter gene analysis.Finally,to further investigate whether the cellular function of miR-124-3p is mediated by the expression of IGF2BP1,rescue experiments were performedResuItsBio informatics results showed that miR-124-3p was down-regulated in EC tissues,and miR-124-3p was expressed at lower levels in patients with high-grade,different histological or menopausal status.Functional assay results showed that overexpression of miR-124-3p reduced cell proliferation,migration and invasion.In contrast,miR-124-3p silencing had the opposite effect.IGF2BP1 is a direct target of miR-124-3p and is negatively regulated by miR-124-3p.Reduction of IGF2BP1 mRNA expression inhibited EC cell proliferation,migration and invasion,and reversed the effects of miR-124-3p on EC cellsConclusionsThe results of this study suggest that miR-124-3p may be a tumor suppressor gene and a potential therapeutic target for treating EC.
