ObjectiveDiabetes and vitamin D deficiency are both common public health problems.Vitamin D is a necessary substance for glucose to stimulate insulin secretion and maintain normal glucose tolerance.It plays an active role in early phase insulin secretion and peripheral insulin action.Vitamin D supplementation is considered a potential and inexpensive diabetes treatment that could not only reduce risk but also improve blood sugar in patients with Type 2 Diabetes Mellitus(T2DM).But there has been less research on the link between vitamin D2 and diabetes.Therefore,this study established an accurate method for quantification of plasma vitamin D metabolites to explore the relationship between plasma vitamin D2 in patients with T2 DM and non-T2 DM patients.MethodsIn this study,liquid chromatography-tandem mass spectrometry(LC-MS/MS)was used to develop analytical methods for 25(OH)D2 and25(OH)D3,the stable metabolites of vitamin D metabolized by liver,and the stable isotope-labeled compounds were selected as internal standard,which could effectively correct the matrix effect in plasma.The methodology was verified according to the internationally recognized CLSI-62 A standard.The electronic medical record data of 797 hospitalized patients were screened,and a total of 596 patients were included after screening by inclusion and exclusion criteria.Patients were divided into diabetic and non-diabetic groups based on clinical information.Plasma vitamin D2 content was determined by LC-MS/MS method.According to the results of plasma vitamin D2 determination,Wilcoxon rank sum test was used to analyze the difference between groups,and logistic regression model was used to find the corresponding influencing factors.ResultsOf the 596 hospitalized patients,138(23.15%)were diagnosed with T2 DM.The Wilcoxon test showed no statistically significant difference in plasma vitamin D2 concentrations between the T2 DM and non-T2 DM groups(p=0.833).After adjustment for confounders by multivariate logistic regression,there was still no significant difference in plasma vitamin D2 concentrations between the two groups(P=0.316,OR: 1.15(0.88,1.49)).The uncorrelated relationship between vitamin D2 and T2 DM was not found to change after incorporating 12 indicators,including demographic characteristics,laboratory indicators and complications,into the logistic regression model by 3 steps,even the OR(1.08(0.9,1.26))did not change in the 3 models.ConclusionsA quantitative measurement method of vitamin D content based on LC-MS/MS was established and verified by methodology,and the reliability of the method was verified.In a cross-sectional study of vitamin D and diabetes,we found no association between vitamin D2 and type 2 diabetes,and there was no significant change in vitamin D2 levels in patients with type 2 diabetes compared to those without type 2 diabetes.Caution should be exercised when supplementing vitamin D2.
