| Objective:To evaluate the curative effect of Wuzang Tongtiao prescription on mild and moderate ulcerative colitis,and the mechanism of its effect was explored by using the network pharmacology method of TCM compound.Methods:1.Clinical efficacy studyAccording to certain criteria,62 cases were included and randomly assigned to treatment and control groups,with 31 cases in each group.The control group was given enteric-coated mesalazine tablets,and the treatment group was treated on the basis of Mesalazine enteric-coated tablets combined with Wuzangtongtiao prescription,which lasted for 4weeks.Statistical analysis was conducted after relevant data were obtained,and the modified Mayo score,total and individual score of TCM syndromes,total clinical effect and efficacy of TCM syndromes before and after treatment were compared between the two groups,and adverse reactions during treatment were observed to evaluate the clinical efficacy and safety of the two treatment programs.2.Network pharmacology researchThrough the network pharmacology of Chinese herbal compound,the main active ingredients,action pathways,biological processes and potential targets of Wuzangtongtiao prescription in the treatment of ulcerative colitis were identified.Results:1.Clinical efficacy studyThe results showed that the treatment group was superior to the control group in Mayo score,total and individual score of TCM syndromes as well as efficacy of TCM syndromes.In terms of total clinical effect,there were no statistically significant differences between the two groups.In the course of treatment,there were no apparent negative reactions in either group,both of which had good safety.2.Network pharmacology researchFinally,152 active ingredients and 230 drug targets of Wuzangtongtiao prescription were obtained,and 1592 disease targets were yielded,135 common targets were got by their interaction.According to the results of protein interaction and target regulatory network,the core targets of Wuzangtongtiao prescription in the treatment of ulcerative colitis may include inflammatory cytokines TNF,IL-6,PTGS2,and PPARG.Apoptosis related factors CASP3,AKT1 and MAPK3,tumor suppressor genes PTEN and TP53,epidermal growth factor receptor EGFR and vascular endothelial growth factor receptor VEGFA.A total of 782 items were obtained by GO functional annotation analysis,including oxidative demethylation,negative regulation of smooth muscle cell migration,leukocyte migration involved in inflammatory response,macrophage activation involved in immune response,toll-like receptor 4 signaling pathway,negative regulation of epithelial cell apoptosis and other biological processes,aminergic synapses,cell apical,endosomal membrane,microvilli,basolateral plasma membrane,serine endopeptidase complex and other cell components,metalloproteinase activity,REDOX enzyme activity,transcriptional inhibitory factor activity,RNA polymerase II transcriptional regulatory region sequence specific binding,threonine kinase inhibitor activity,5-hydroxytryptamine binding,MDM2/MDM4 family protein binding and other molecular functions.A total of 174 items were obtained by KEGG enrichment analysis,including caffeine metabolism,steroid hormone biosynthesis,lipolysis regulation,glutathone metabolism,colorectal cancer pathway,JAK-STAT signaling pathway,Nod-like receptor signaling pathway,VEGF signaling pathway,NFk B signaling pathway,etc.Conclusion:Wuzangtongtiao prescription has obvious clinical efficacy in the treatment of patients with mild to moderate ulcerative colitis,can significantly relieve the symptoms,and the safety is relatively ideal,which is a new idea for drug selection in the treatment of ulcerative colitis.The process of its therapeutic effect may be related to the active components such as quercetin,kaempferol and luteolin,which regulate glutathione metabolism,colorectal cancer,JAK-STAT signaling pathway,act on TNF,CASP3,PTEN,EGFR,AKT1,TP53 and other targets,affects cell cycle,poptosis and inhibits inflammation. |
