| Objective:To investigate the expression differences of S100A6,β-catenin,GST-π,ToPoⅡ and other factors in human gastric cancer cells during the pathological phase of tumor.Methods:Clinical data of 128 patients who underwent gastric cancer resection in our hospital from June 2020 to June 2022 were retrospectively analyzed,including general data,postoperative pathological specimens and pathological examination results.According to the inclusion and exclusion conditions,87 subjects were selected.Of the87 patients,62 had lymph node metastasis and 25 had no lymph node metastasis.Postoperative pathological stages were T2 stage 20(23.0%),T3 stage 22(25.3%),T4 a stage 24(27.6%),and T4 b stage 21(24.1%).Pathological samples of primary gastric cancer and lymph nodes were decomposed,RNA was extracted and reverse-transcribed into c DNA,target molecular primers were designed for PCR,dissolution curves were constructed,and mRNA expression levels were read out.SPSS 22.0 statistical software was used to analyze the differences in patients’ general situation,histopathological grade,expression levels of multidrug resistance related factors among groups T2,T3,T4 a,T4b,lymph node metastasis and non-metastasis,and lymph node metastasis.Results:There were no significant differences in S100A6,β-catenin,GST-π and ToPoⅡ among male and female groups.There were no significant differences in S100A6,β-catenin,GST-π and ToPoⅡ among age groups.The mRNA expression of patients in the highly differentiated,moderately differentiated,poorly differentiated and undifferentiated groups of postoperative pathological tumor histological grading was in line with normal distribution.The results of one-way analysis of variance showed that F was 134.987,158.856,132.987 and 148.523,respectively.The P values were 0.117,0.027,0.147 and 0.171.There was no statistical significance in S100A6,GST-π and ToPoⅡ group,and no postmortem analysis was necessary.The mRNA expression of β-catenin was statistically different.Further pound-wise comparison showed that the expression of β-catenin mRNA in the highly differentiated group was significantly lower than that in the moderately differentiated,poorly differentiated and undifferentiated groups(P= 0.024,0.006),and the moderately differentiated group was significantly lower than that in the poorly differentiated and undifferentiated groups(P=0.021).The mRNA expressions of S100A6,β-catenin,GST-π and ToPoⅡ in patients with T2,T3,T4 a and T4 b stages were in line with normal distribution.The results of one-way analysis of variance showed that F were 160.987,175.462,155.675 and182.276,respectively.All P values were <0.001,indicating that mRNA expression of at least two groups was statistically different.Further pin-wise comparison showed that S100A6,β-catenin,GST-π and ToPoⅡ had significant differences in the four T fractions,showing a linear increasing trend.The mRNA expressions of S100A6,β-catenin,GST-π and ToPoⅡ in the group without lymph node metastasis and the group with lymph node metastasis were in line with normal distribution.The results showed that the mRNA level of S100A6 in lymph node metastasis group was significantly higher than that in no lymph node metastasis group(1.56±0.37 vs 3.78±0.26,t=1.56,<0.001).The mRNA level of β-catenin in lymph node metastasis group was significantly higher than that without lymph node metastasis group(1.16±0.19 vs 2.28±0.25,t=-2.86,<0.001).The mRNA level of GST-π in lymph node metastasis group was significantly higher than that without lymph node metastasis group(2.12±0.15 vs 3.51±0.26,t=1.16,<0.001).The mRNA level of ToPoⅡ in lymph node metastasis group was significantly higher than that without lymph node metastasis group(1.12±0.19 vs 2.47±0.23,t=0.42,<0.001).Conclusions:1.The expression levels of S100A6,β-catenin,GST-π and ToPoⅡ were not correlated with age and gender.2.The expression level of multidrug resistance factor β-catenin is about higher in gastric adenocarcinoma with lower histological differentiation,which may be one of the factors aggravating chemotherapy resistance of gastric adenocarcinoma with lower differentiation.3.With the increase of tumor invasion depth,the expression levels of S100A6,β-catenin,GST-π and ToPoⅡ also showed an upward trend.4.With the occurrence of local lymph node metastasis,the expression levels of S100A6,β-catenin,GST-π and ToPoⅡ also showed an upward trend. |
