| Objective: This study aimed to establish a rat cardiac arrest/cardiopulmonary resuscitation(CA/CPR)model and intervene by reference injection to observe whether the attached injection has a protective effect on rat cardiomyocytes after CA/CPR,and to observe its protective mechanism and whether to improve CA/CPR rat myocardial dysfunction by regulating autophagy by regulating the PI3K/Akt pathway.Methods: By selecting 40 healthy adult SD rats for CA/CPR modeling,the rats were finally divided into sham surgery group,normal resuscitation group,2ml/kg group,and 7ml/kg group.After all rats were adaptively fed,30 of the rats were given asphyxiation combined with potassium glacial chloride to induce rat cardiac arrest,the rats were recorded,and when the arrest was 5minutes later,routine cardiopulmonary resuscitation and other measures were immediately given to rescue,and after the spontaneous circulation of the rats was restored,they were randomly divided into normal resuscitation group,2ml/kg group,and 7ml/kg group,and 1 h after rat resuscitation,9 to 5 am the next day,1 ml / time with normal saline,2 ml / kg / time,and 7 ml / kg /Times.The remaining 10 were used as sham surgery groups,weighed 1 h,the next morning 9 and 5 pm were injected with 1ml of normal saline.ELISA measured serum creatine kinase isoenzyme(CK-MB)and cardiac troponin I(c Tn I)levels 24 hours after the intervention,and measured the degree of myocardial injury in rats after cardiopulmonary resuscitation in cardiac arrest.HE detects the manifestations of pathological changes in the structure of the myocardium after cardiac arrest;The Western Blot method detected the expression levels of autophagy-related proteins LC3 II.,Beclin-1,PI3 K and Akt pathway proteins.Results: 1.Compared with the Sham Operation Group,he normal resuscitation group showed severe myocardial damage,irregular arrangement of myocardial cells,disruption of myocardial fibers,and an increase in the number of blood HE stain between the myocardium,there is a lot of inflammatory cell infiltration between the myocardium.The myocardial injury in Shenfu intervention group was milder than that in normal resuscitation group,and the milder in Shenfu 7ml/kg group.2.Compared with the sham group,serum CK-MB and c Tn I expression values were increased to varying degrees(P<0.05)in both resuscitated and injected rats.The expression values of CK-MB and c Tn I in the normal resuscitation group were higher than those in the sham group(P<0.05),the CK-MB and c Tn I levels in the 2 ml/kg injection group with 2 ml/kg injection were lower than those in the normal resuscitation group(P<0.05),while the CK-MB and c Tn I levels(P<0.05)in the 7 ml/kg injection enrichment group were lower and more significantly lower than in the normal resuscitation group.The results showed that the myocardial function of rats in the normal resuscitation group was significantly impaired,and the cardiac function of rats could be significantly improved after CA/CPR injection group,which proved that the effect of high doses was more obvious..3.Western blotting showed that LC3 II and backlin-1 were expressed in all groups.LC3 II and beclin-1 expression increased in the normal resuscitation group,and PI3 K and Akt expression decreased(P<0.05).Compared with the normal resuscitation group,the expression of LC3 II and beclin-1 was reduced in the Shenfu group,while the expression of PI3 K and Akt at 2 ml/kg increased(P<0.05).The expression of LC3 II and beclin-1decreased in the 7 ml/kg and normal resuscitation groups,and the expression of PI3 K and Akt increased(P<0.05).The results showed that myocardial hyperphagy occurred after CA/CPR,and Shenfu injection could inhibit the level of autophagy,upregulate the PI3K/Akt signaling pathway,and relieve myocardial injury.Conclusion:(1)The attached injection can improve myocardial dysfunction after cardiac arrest/cardiopulmonary resuscitation in rats.(2)After cardiac arrest/cardiopulmonary resuscitation,rat myocardial injury was significantly improved after intervention in the attached injection group,indicating that excessive autophagy was an important mechanism leading to myocardial injury.(3)The attached injection can inhibit excessive autophagy by upregulating the PI3K/Akt signaling pathway,thereby exerting the myocardial protective effect after cardiac arrest/cardiopulmonary resuscitation,and the effect of the high-dose group is more obvious. |
