Objective:Taking three kinds of Alpinia,which are related to the galangal,the fructus galangae and the galanga,as the research object,the flavonoids of the three Alpinia are extracted and prepared.The model of gastric ulcer cold syndrome was replicated in rats to study the effects of three flavonoids from Alpinia spp.on the intestinal flora and short chain fatty acids(SCFAs)of rats with gastric ulcer cold syndrome.To clarify the relationship between the anti-gastric ulcer cold syndrome effect of the three-flavored Chinese medicine of the genus Alpinia and the micro-ecological regulation,and to explore the correlation between"physical relationship-Chinese medicine-micro-ecology-regulation".Methods:(1)Extract the flavonoids of the three-flavored Alpinia Chinese medicine by the ethanol reflux extraction method;purify the flavonoids by the alkaline solution and acid precipitation method.(2)The rats were intragastrically administered with 4℃Bingzhimu water decoction and 15%glacial acetic acid aqueous solution to replicate the rat model of gastric ulcer cold syndrome.Fresh feces of each group of rats were collected with sterile tweezers.Bacterial genes are sequenced,and basic quality control analysis,OTU cluster analysis,Alpha diversity analysis,Beta diversity analysis,and bacterial flora composition and difference analysis are performed on the results using the Omicsmart Metagenome platform.(3)The contents of acetic acid,propionic acid,isobutyric acid,butyric acid,isovaleric acid,and valeric acid in rat feces were measured by a gas-mass spectromete.Results:(1)The combination of quality control analysis,OTU analysis and intestinal flora diversity analysis shows that the experimental data processing process is strict,the sample sequencing quality is good,it has a certain depth and representativeness,and the species uniformity and richness are good.The flavonoids of 3 kinds of Chinese medicine of the genus Alpinia can promote the recovery of the abundance and diversity of the intestinal flora in rats with gastric ulcer cold syndrome.The structure of the flora in each administration group is highly similar,and the fructus galangae group and the galanga group The similarity of the group flora structure is higher.(2)The composition and difference analysis of the intestinal flora showed that there were significant differences in the intestinal flora between the galangal group,the fructus galangae group,the galanga group and the model group.The main manifestations are the decrease of the abundance ratio(ie F/B value)of Firmicutes and Bacteroides,and the significant increase of beneficial bacteria such as Lactobacillus,Prevotella,Clostridium,etc.(p<0.01 or p<0.05),Conditional pathogenic bacteria such as Cyanobacteria,Proteobacteria,Escherichia-Shigella,etc.were significantly reduced(p<0.01 or p<0.05).(3)LEfSe analysis shows that,compared with the blank group,Pasteurella,Enterobacter,Escherichia-Shigella,Rotabacterium,Morganella morganii and Pasteurella in the model group increased significantly(p<0.01),and Bifidobacterium Significant decrease(p<0.01);Compared with the model group,Pasteurella and Rotabacterium decreased significantly in the fructus galangae group(p<0.01),Bacillus,Bifidobacterium,and Lactobacillus increased significantly(p<0.01);The segmental filamentous bacteria of galanga species increased significantly(p<0.01),and there was no significant difference between the galangal group and the model group.(4)The linear ranges of acetic acid,propionic acid,isobutyric acid,butyric acid,isovaleric acid,and valeric acid are respectively11.91~789.26μg/m L(R~2=0.9976),15.98~782.55μg/m L(R~2=0.9979),0.09~394.31μg/m L(R~2=0.9986),0.06~394.40μg/m L(R~2=0.9986),0.04~395.52μg/m L(R~2=0.9979),0.02~393.36μg/m L(R~2=0.9982),the average recovery rate is 85.46 Within the range of%~102.59%,RSD<2.88(n=6).(5)Compared with normal rats,the SCFAs content of gastric ulcer cold syndrome rats were significantly decreased(p<0.01);after drug intervention,compared with the model group,the SCFAs content of the galangal group increased although not statistically significant;fructus galangae The content of SCFAs in the group increased significantly(p<0.05,p<0.01);The content of acetic acid,propionic acid,isobutyric acid,isovaleric acid,and valeric acid in the galanga group increased significantly(p<0.05,p<0.01),but the butyric acid content increased but not statistically significant.Conclusion:(1)The influence of the flavonoids of 3 kinds of Chinese medicines of the genus Alpinia on the intestinal flora of rats with gastric ulcer cold syndrome is related to the genetic relationship,and the genetic relationship between the fructus galangae and the galanga is more closely related.(2)The flavonoids of 3 kinds of Chinese medicine of the genus Alpinia can improve the disorder of intestinal flora in rats with gastric ulcer cold syndrome,increase the abundance and diversity of intestinal flora,reduce the abundance of pathogenic bacteria,and increase the abundance of beneficial bacteria.Degree;the indicator species that cause the cold syndrome of gastric ulcer may be Pasteurella,Enterobacter,Escherichia-Shigella,Rotabacterium,Morganella morganii and Pasteurella,which play a role in resisting the cold syndrome of gastric ulcer The indicator species may be Prevotella,Bifidobacterium,Bacillus,Lactobacillus and segmented filamentous bacteria.(3)The GC-MS method can quickly and accurately detect the content of SCFAs in the intestinal tract;The flavonoids of 3 kinds of Chinese medicine of the genus Alpinia can play a role in resisting the cold syndrome of gastric ulcer by increasing the content of SCFAs,which are metabolites of intestinal flora.
