Objective:Taxilli Herba is a semi-parasitic plant with a wide range of host sources.In this paper,the cold and hot medicinal properties were compared by using Taxilli Herba on the hosts of the flat medicinal mulberry(called Taxilli Herba parasitizing mulberry),on the hosts of the warm medicinal cinnamon(called Taxilli Herba parasitizing cinnamon)and on the hosts of the cold medicinal willow(called Taxilli Herba parasitizing willow).and establish a set of methods for detection and evaluation of Taxilli Herba properties based on the influence of the host;Through the detection of the material composition and content of Taxilli Herba and its host plants from different host sources,the possible mechanism of the host affecting the quality of Taxilli Herba medicinal materials is explored from the material composition point of view,and the host’s understanding of the use of Taxilli Herba medicinal materials is improved.Provide a reference basis for the clinical rational use of drugs.Methods:1.Microcalorimetry was used to study the medicinal properties of Taxilli Herba parasitizing mulberry,Taxilli Herba parasitizing cinnamon and Taxilli Herba parasitizing willow.The cold and hot medicinal properties of Taxilli Herba parasitizing mulberry,Taxilli Herba parasitizing cinnamon and Taxilli Herba parasitizing willow materials were evaluated by isothermal microcalorimeter and Escherichia coli(E.coli).In the experiment,the first peak time,the first peak power,the second peak time,the second peak power and enthalpy were used as the evaluation indexes to evaluate the cold and hot medicinal properties of Taxilli Herba parasitizing mulberry,Taxilli Herba parasitizing cinnamon and Taxilli Herba parasitizing willow.2.Using the cold and hot plate differential method to study the medicinal properties of Taxilli Herba parasitizing mulberry,Taxilli Herba parasitizing cinnamon and Taxilli Herba parasitizing willow.Detection of Kidney Yin Deficiency Mice by Animal Behavior Intelligent Monitoring System.The cold and hot medicinal properties of Taxilli Herba parasitizing mulberry,Taxilli Herba parasitizing cinnamon and Taxilli Herba parasitizing willow were evaluated by tracking the weight,volume of water intake,food intake,oxygen consumption and temperature tendency of mice with Kidney Yin Deficiency,combining with the c AMP/c GMP ratio in plasma and the activity evaluation of Na~+K~+-ATPase and SOD in liver tissue.3.Metabolomics was used to detect the material components of Taxilli Herba parasitizing mulberry,Taxilli Herba parasitizing cinnamon,Taxilli Herba parasitizing willow and their host plants mulberry branch,cinnamon branch,willow branch.In the experiment,UPLC-MS/MS was used to separate and detect the material components of the samples,and the metabolites of Taxilli Herba from three different host sources and their host plants were analyzed qualitatively and quantitatively by mass spectrometry.At the same time,the method of multivariate statistical analysis was used to screen the different metabolites between Taxilli Herba and its host by using PCA,cluster analysis and OPLS-DA.Results:1.It was found that Taxilli Herba parasitizing mulberry and Taxilli Herba parasitizing willow could inhibit the growth and metabolism of E.coli in varying degrees with the increase of drug concentration.The second peak power,second peak time,growth rate constant and enthalpy of xilli Herba parasitizing mulberry and Taxilli Herba parasitizing willow decreased with the increase of drug concentration.It was found that Taxilli Herba parasitizing cinnamon could promote the growth and metabolism of E.coli in varying degrees with the increase of drug concentration,mainly in the growth rate constant,the second peak power and enthalpy.2.Using hot and cold plate differential method to test kidney yin deficiency mice.In the change of weight growth,the weight growth of mice in each high-dose group was in a low state,and the weight growth trend of mice in each low-dose group was fast.The influence of different host parasitisms on the weight growth of mice was shown as follows:Taxilli Herba parasitizing willow group>Taxilli Herba parasitizing mulberry group.>Taxilli Herba parasitizing cinnamon group.The water intake and food intake of mice were changed with the change of weight of mice in each group.The increase of body weight of mice was accompanied by the increase of water intake and food intake.On the contrary,it was more obvious in the low dose group of Taxilli Herba parasitizing willow,and there was a corresponding trend of increase.Compared with the oxygen consumption of mice in each group,Taxilli Herba parasitizing cinnamon high dose group and Taxilli Herba parasitizing mulberry high dose group can reduce the trend of oxygen consumption in mice.In the comparison of the proportion of stay in the warm zone,the proportion of stay in the hot zone will continue to decrease with the increase of modeling days in the model group and each administration group,and the difference between each administration group is not obvious.In comparison of Na~+-K~+-ATPase e activity,there were significant differences among low dose groups.The SOD activity in the low-dose group of Taxilli Herba parasitizing willow increased,and Taxilli Herba parasitizing willow low dose group>Taxilli Herba parasitizing mulberry low dose group>Taxilli Herba parasitizing cinnamon low dose group;The c AMP/c GMP ratio of mice in the Taxilli Herba parasitizing mulberry low dose group low-dose group was decreased,which was significantly different from that in the model group.3.By UPLC-MS/MS detection platform and database MWDB,409 metabolites and 74 characteristic metabolites were detected in mulberry branch;386metabolites and 48 characteristic metabolites were detected in cinnamon branch;441 metabolites and 66 characteristic metabolites were detected in willow branch;441 metabolites and 23 characteristic metabolites were detected in Taxilli Herba parasitizing Mulberry;440 metabolites and 20 characteristic metabolites were detected in Taxilli Herba parasitizing Cinnamon;423metabolites and 7 characteristic metabolite were detected in Taxilli Herba parasitizing Willow;12 characteristic metabolites shared by mulberry branch and Taxilli Herba parasitizing mulberry;8 characteristic metabolites shared by cinnamon and Taxilli Herba parasitizing cinnamon;4 characteristic metabolites shared by willow and Taxilli Herba parasitizing willow.Combining the analysis of the characteristic material components and differential metabolites of Taxilli Herba parasitizing mulberry,Taxilli Herba parasitizing cinnamon and Taxilli Herba parasitizing willow,the main material components that distinguish between Taxilli Herba parasitizing mulberry,Taxilli Herba parasitizing cinnamon and Taxilli Herba parasitizing willow were screened.Among them,7 species of Taxilli Herba parasitizing mulberry,7 species of Taxilli Herba parasitizing cinnamon,and and 5 species of Taxilli Herba parasitizing willow.Conclusions:1.According to the results of microcalorimetric method and cold-hot plate differential method,the medicinal properties of Taxilli Herba parasitizing cinnamon may be warm-hot medicinal properties,and Taxilli Herba parasitizing willow may be cold medicinal properties.The cold and hot medicinal properties of Taxilli Herba parasitizing mulberry are between Taxilli Herba parasitizing mulberry willow and Taxilli Herba parasitizing mulberry cinnamon.Combining the experimental results and the description of the medicinal properties of Taxilli Herba parasitizing mulberry in previous generations of herbal medicine,it is inferred that the medicinal properties of mulberry parasitism may be flat or cool.2.Metabolomics technology was used to analyze the material components between Taxilli Herba and host from different host sources.It was found that there was a transmission of material components between Taxilli Herba and hosts,and the material components and contents transmitted to Taxilli Herba were also different in different hosts.The components and contents of Taxilli Herba parasitizing mulberry,Taxilli Herba parasitizing cinnamon and Taxilli Herba parasitizing willow parasitism were analyzed.It was found that there were differences in the components and contents of Taxilli Herba from three different host sources,which might be the main reason for the differences in the quality and medicinal properties of medicinal materials between Taxilli Herba parasitizing mulberry,Taxilli Herba parasitizing cinnamon parasitism and Taxilli Herba parasitizing willow The experiment included the host into the scope of the research on the quality of Taxilli Herba medicinal materials,and provided reference for the clinical rational use and development and utilization of Taxilli Herba medicinal materials from different hosts.
