Objective: 1.Based on data mining technology,summarize the clinical medication rules of Professor Hu Yueqiang in the diagnosis and treatment of insomnia with kidney Yang deficiency,summarize the drug characteristics,drug frequency and drug combination rules,and discuss and confirm the concept of diagnosis and treatment center.2.Through network pharmacology,the effective ingredients and target sites of core drugs for insomnia were discussed and the signal pathway was detected and analyzed,and the relevant "core drug pair-component-target" action network and PPI network were constructed,and the GO functional enrichment analysis and KEGG pathway enrichment analysis were carried out to predict the potential targets and signal pathways of core drugs for insomnia.Methods: 1.Cases of patients with nephroyang deficiency insomnia in Professor Hu Yueqiang’s outpatient Department of Encephalopathy,The First Affiliated Hospital of Guangxi University of Traditional Chinese Medicine from September 2020 to September 2022 shall be collected.The research objects shall strictly meet the inclusion and exclusion criteria,conduct data integration processing,and standardize the name of traditional Chinese medicine.The data were imported into the traditional Chinese medicine inheritance computing platform V3.0 to establish a special database.The frequency analysis of traditional Chinese medicine,the analysis of drug category,sex,taste and normalization,the association rule analysis and cluster analysis of commonly used traditional Chinese medicine,through scientific methods to find the relationship between core drug pairs and drug compatibility,to explore the clinical medication law of Professor Hu Yueqiang’s diagnosis and treatment of this disease.2.Detect core components of drugs and drug-related targets through TCMSP database,search targets related to insomnia using Gene Cards and Drug Bank disease database to obtain targets of mutual interaction between the two,and conduct interaction analysis on preprotein of targets with mutual interaction in Strings database.GO function enrichment analysis and KEGG pathway enrichment analysis were carried out by R language.Results: 1.202 prescriptions were recorded in this study,involving a total of 109 traditional Chinese medicines,among which the top 10 drugs with frequent frequency were aconite,glycyrrhiza,oyster,keel,Phellodendrin,sand nut,tortoisesia,Fu Shen,sour jujube nut and codonopsis.The top 5 drugs in terms of efficacy were: tonifying deficiency,clearing heat,warming Li,calming liver and extinguishing wind,and dispelling dampness.The four qi in prescription from high to low are: cold,warm,flat,hot,cool;The five tastes were sweet,hot,bitter,salty and sour in order from high to low.The top 5 drugs in the frequency of meridian use were heart channel,kidney channel,spleen channel,liver channel and lung channel.Drug cluster analysis was divided into three categories: "Aconite,sand kernel,Yellow cypress,keel,oyster","Codonopsis,Aconite,licorice,oyster,yellow Cypress","Fu Shen,stone calamus,licorice,keel,oyster";Drug association rule:Aconiti aconiti and Phelloderma phelloderma had the highest correlation degree in prescriptions(120 times),and this drug pair was also the core drug pair with the highest frequency of treating kidney Yang deficiency type insomnia.2.The network pharmacological study found that the core drug pair contained 30 kinds of active ingredients and 179 drug related targets,among which the main active ingredients were quercetin,tetrahydroafrhexine,tetrahydroberberine,β-sitosterol,coryneine,etc.A total of 367 disease-related targets were screened,and there were 32 intersection targets between the core drug pair and the disease.The key targets were IL6,CASP3,IL1 B,VEGFA,TNF,IL10,NR3C1,HIF1 A,SLC6A4,AHR,etc.GO enrichment analysis obtained 1452 biological processes(BP),involving the response to lipopolysaccharide,molecular function of response to foreign biological stimuli,angiogenesis regulation and other biological processes.Twenty-seven cell components(CC)were obtained,which involved the organic components of postsynaptic membrane,plasma membrane raft,neuronal projection cytoplasm,etc.A total of 117 molecular functions(MFS)were obtained,including binding to cytokine receptors,activity of neurotransmitter receptors,and activity of postsynaptic neurotransmitter receptors.A total of 86 pathways were identified by KEGG enrichment analysis,and the anti-inflammatory and anti-apoptotic effects are mainly played through IL-17 signaling pathway,tumor necrosis factor signaling pathway,apoptosis,lipid and arteriosclerosis,and Cytokinecytokine receptor interaction and other signaling pathways.Conclusion: 1.The medicines used in TCM treatment of kidney Yang deficiency type insomnia are mainly deficiency tonifying drugs and heat-clearing drugs;It is related to the heart,kidney and spleen channels,and its main pathogenesis may be the deficiency of kidney Yang,the deficiency of Yang and the abundance of Yin.If Yin is too abundant,Yin and Yang will be denied and Yang will not enter Yin,which leads to the floating of deficiency Yang,disturbing the mind and sleepless at night.In the analysis of drug association,aconite and yellow cypress ranked the first.Yellow cypress sijunhuo,clear the heat of empty fire,both use together,lead phase fire return,true Yang can be supplemented,empty fire can clear diarrhea,reflecting Professor Hu Yueqiang’s main diagnosis and treatment of kidney Yang deficiency type insomnia.And "Aconitum-Phelloderma" is Professor Hu Yueqiang’s treatment of kidney Yang deficiency type insomnia core drug pair.2.Through network pharmacology,it was found that the core drug pair acts on the main core targets such as IL6,CASP3,IL1 B,VEGFA,TNF,IL10,NR3C1,HIF1 A,SLC6A4 and AHR through quercetin,tetrahydroafribohexine,tetrahydroberberine,β-sitosterol,corylanine and other active ingredients.It involves many biological processes,such as the reaction to lipopolysaccharide,the function of molecular reaction to foreign biological stimuli,and the regulation of angiogenesis.Multiple signaling pathways such as AGE-RAGE signaling pathway,IL-17 signaling pathway,TNF signaling pathway,apoptosis,interaction between lipids and arteriosclerosis,and Cytokine-cytokine receptor are regulated to exert anti-inflammatory and anti-apoptosis functions in the treatment of insomnia.
