Objective:In this study,MAX dimerization Protein 1(MXD1)gene olymorphism was used as the main object to investigate its association with Ischemic Stroke(IS)and its clinical and biochemical indices,taking into account the basic theoretical knowledge of Chinese medicine.Methods:This study included 774 patients with ischemic stroke in the case group and 793 control samples,a total of 1567 cases.According to the Trial of the Diagnostic Criteria for Syndrome Differentiation of Stroke,ischemic patients were differentiated.The Polymorphic sites rs12475412 and rs7349311were genotyped by Massarray technique.The STAGO fully automated anticoagulant coagulometer and the Hitachi 00-020ISE fully automated biochemical analyzer were used,respectively,and the determination of serum coagulation function and blood glucose levels in patients with ischemic stroke was performed according to standardized laboratory procedures.Genetic association analysis was performed by PLINK software,and other related analysis was performed by SPSS software.Results:1.The test of Hardy-Weinberg Equilibrium(HWE)The genotype frequency distribution of the MXD1 gene rs7349311 (PHWE=0.074)and rs12475412(PHWE=0.871)loci in the control group were in accordance with Harwin equilibrium.2.Genotype frequency distribution:There was no statistically significant difference in the distribution of genotype frequencies of the MXD1 gene rs7349311 and rs12475412 loci between the two groups of stroke cases and controls(all P>0.05).Between the pathology and control groups,the P values for both MXD1 loci for wind,fire-heat,phlegm,blood stasis and qi deficiency evidence were also all>0.05,with no statistically significant differences.3.Association study of MXD1 gene Polymor Phism with IS and susceptibility to TCM syndromes:The MXD1 rs7349311 and rs12475412 gene polymorphisms were not associated with the risk of IS(P>0.05 for all),and polymorphisms at both MXD1 loci were not associated with the risk of IS(P>0.05 for all groups).4.Correlation between MXD1 gene polymorphism and TCM of IS patients:The rs7349311 polymorphism of MXD1 gene is significantly associated with the risk of blood stasis syndrome in Han female patients with ischemic stroke,and significantly associated with the risk of Qi deficiency syndrome in Han female ischemic stroke patients.The rs12475412 polymorphism of MXD1gene is significantly associated with the risk of blood stasis syndrome in Han female patients with ischemic stroke,and significantly associated with the risk of Qi deficiency syndrome in Han female ischemic stroke patients(P>0.05 for all).5.Correlation of MXD1 gene polymorphisms with clinical and biochemical indicators in patients with IS:(1)The rs12475412 polymorphism(MXD1 gene)was significantly associated with FPG levels in patients with ischaemic stroke,which(recessive model:Padj=0.037);The rs12475412 polymorphism was significantly associated with TG levels in IS patients(dominant model:Padj=0.034);The rs7349311 Polymorphism was significantly correlated with the PTA level of IS patients(recessive model:Padj=0.027);The correlation between rs7349311 Polymorphism and APO-B(additive model:Padj=0.007;dominant model:Padj=0.003)level,LDL-C(additive model:Padj=0.043;dominant model:Padj=0.010)level,TC(additive model:Padj=0.047;dominant model:Padj=0.013)level,TG(additive model:Padj=0.022;dominant model:Padj=0.011)level of IS patients was statistically significant.(2)There was no significant correlation between the Polymorphisms of MXD1’rs7349311 and rs12475412 gene and SBP and DBP of IS patients(P>0.05);(3)There was no significant correlation between the rs7349311 Polymorphism of MXD1 gene and the levels of FPG and P2h PG(P>0.05);(4)The rs12475412 polymorphism of the MXD1 gene did not correlate with APTT,FIB,INR,D-D,PLT,PT,PTA,and also TT levels in patients with IS(P>0.05);(5)There was no significant correlation between the Polymorphisms of rs7349311 and rs12475412 of MXD1 gene and CRP,Hcy and UA of IS Patients(all P>0.05).Conclusion:(1)The rs7349311 and rs12475412 polymorphisms in the MXD1 gene may affect the occurrence of qi deficiency and blood stasis syndrome in patients with ischemic stroke.(2)The rs12475412 Polymorphism of MXD1 gene may affect the blood glucose level and lipid metabolism of Patients with ischemic stroke.(3)The MXD1 gene rs7349311 polymorphism may affect coagulation and lipid metabolism in patients with ischemic stroke.
