| Objective: In this study,a variety of biological information network databases(such as TCMSP,HERB,Gene Cards,etc.)and related literature were deeply explored to study the network pharmacology of Anluo Huayan pills in the treatment of Hepatic fibrosis.Methods: Through the investigation of TCMSP,HERB and other databases and related literature,the active components of our prescription were obtained,and the target gene was reverse matched on TCMSP,Swiss ADME database and Swiss Target Prediction database were used to make target prediction,and the active components that did not meet the conditions and did not predict the corresponding target were eliminated.The duplicated data were deleted,and the effective chemical components and gene targets of 14 herbs in Anluo Huaxian Pills were finally obtained.The gene names of the obtained targets were standardized by using Unipro database.At the same time,gene targets related to Hepatic fibrosis were retrieved in Gene Cards database,and the two groups of genes were mapped to obtain the intersection target genes,which could be used as the potential target for the treatment of Hepatic fibrosis.Furthermore,String database and Cytoscape 3.9.1 software were used to analyze potential targets and construct the protein interaction network diagram,while screening out the core targets of Anluo Huayan pills for the treatment of Hepatic fibrosis.The intersection genes of Anluo Huayan pills and Hepatic fibrosis were input into Metascape database for enrichment analysis of GO function and KEGG pathway,to obtain related biological processes and signaling pathways,and to draw bubble maps of corresponding pathways.Target genes with the top 3 degrees in PPI network and active components with the number of target genes at the top 3 were selected for molecular docking and verification.Results: Through TCMSP,HERB database and literature search,125 active components of Anluo Huayan pills and 209 target genes of Hepatic fibrosis were obtained.Ten core compounds were quercetin,luteolin,kaempferol,linolenic acid,linoleic acid,hirudin,methyl ferulic acid,arachidonic acid,eicosapentaenoic acid,naringin.The 20 core targets were ALB,TNF,GAPDH,AKT1,IL6,VEGFA,TP53,IL1 B,JUN,MAPK3,EGFR,STAT3,CASP3,MMP9,SRC,CXCL8,HIF1 A,MYC,EGF,ESR1;The core biological processes and pathways may include hormone response,positive regulation of cell movement,cascade regulation of MAPK,inflammatory response,cell response to nitrogen compounds,regulation of cell activation,cell response to lipids,cancer signaling pathway,lipid and atherosclerosis signaling pathway,PI3K-Akt signaling pathway,etc.Conclusion(s):(1)Anluo Huayan pills can treat hepatic fibrosis through synergistic effect of multi-component,multi-target and multi-pathway.(2)The results showed that the main chemical components of Anluo Huaxian pills,such as quercetin,luteolin,kaempferol,linolenic acid and so on,have the effects of antioxidant,anti-inflammatory,anti-tumor,free radical scavenging,anti-fibrosis and liver cell protection.(3)ALB,TNF,GAPDH,IL6,AKT1,VEGFA and TP53 may be the key target genes of Anluo Huayxin pills in the treatment of hepatic fibrosis.(4)Anluo Huayan pills play a role in the treatment and anti-Hepatic fibrosis by regulating cancer pathway,PI3K-Akt signaling pathway,MAPK signaling pathway,hepatitis B,hepatitis C and other signaling pathways. |
