Objective: A novel protein assay was constructed to sensitively detect the content of trace proteins in the serum of patients,and the correlation between inflammatory factor IL-6 of cerebrovascular disease and intracranial and extracranial large artery atherosclerotic stenosis was explored with fluorescence colorimetric dual mode and its clinical application.Methods: Three kinds of hydrolysates and fluorescent probes were used to detect the activity of alkaline phosphatase: APP,Si-FITC NPs and p NPP.The IL-6 protein was detected by two reading modes: colorimetry and fluorescence.The UV absorption value of APP hydrolysate AP and the linear relationship between APP fluorescence intensity and IL-6 protein concentration were determined.The ELISA method was then applied to the detection of clinical samples.Clinical examination methods: A total of 40 cerebral infarction patients were clinically analyzed using whole brain digital subtraction angiography(DSA),and the patients were included in the study based on the presence or absence of intracranial and extracranial large artery atherosclerosis,in order to divide them into two parts,the control group and the stenosis group,and then the stenosis group was divided into mild,moderate,and severe stenosisgroups according to the degree of stenosis in the intracranial and extracranial vessels,respectively.The IL-6 protein content in the serum pretreatment of40 patients was detected using an ELISA method that has been established in experimental methods,and the differences in the clinical data among different groups of patients were compared,and the IL-6 protein content in the serum was analyzed for correlation with the indicators of clinical risk factors related to the degree of cerebral artery stenosis.Results: 1.Based on the APP hydrolysis colorimetric fluorescence dual-mode ELISA to detect IL-6 protein content in complex samples,the experimental results were consistent with the results of clinical examination,and quantitative detection of trace protein was achieved.2.The experimental material for detection is simple,and the experimental method has high specificity as well as high sensitivity,resulting in a two orders of magnitude reduction in the detection limit compared with commercially available IL-6 protein ELISA test kits.3.There is a certain correlation between the level of IL-6 and the atherosclerotic stenosis of intracranial and extracranial great arteries,and the expression of IL-6 is significantly increased in patients with severe atherosclerotic stenosis.4.IL-6 may be a diagnostic or predictive factor for atherosclerotic stenosis of intracranial and extracranial great arteries.Conclusion: There was a close relationship between IL-6 and intracranial and extracranial atherosclerotic stenotic changes,which could reflect the severity of cerebral infarction and make judgment on the stability of arteriosclerotic plaques.The effects and molecular mechanisms of IL-6 onatherosclerotic plaque formation and plaque properties may provide new insights into the prevention and treatment of atherosclerosis,but also indicate new ways to inhibit the inflammatory response,prevent atherosclerosis at an early stage,and delay the progression of atherosclerotic plaque formation.
