Study On The Molecular Mechanism Of Jiawei Qifang Weitong Granules In The Treatment Of Gastric Cancer Based On Cell Ferroptosis

Objective:To investigate the effect of Jiawei Qifang Weitong Granules on gastric cancer cells and the molecular mechanism of inhibiting ferroptosis.Methods:A total of 20 SD male rats were selected,each weighing about 300 g,7-8 weeks old.Modified Qifang Weitong Granules were intervened for 7 days to prepare drug-containing serum.After resuscitation and passage of AGS cells,gastric cancer AGS cells were randomLy divided into blank control group(CK&AGS group),blank serum group and drug-containing serum group.After 2 days of intervention,the cell activity of each group was detected(CCK8 method),the cell morphology was observed by inverted fluorescence microscope,the contents of MDA and GSH were detected by colorimetry,and the expression levels of SLC7A11,P53 and GPX4 were detected by qPCR and Western Blotting.Results:1.The results of CCK8 showed that AGS cells were observed as epithelioid adherent growth under the microscope.The cells in the normal group grew well.Compared with the AGS group,the cells in the blank serum group were observed under the microscope.There was no significant difference between the groups,indicating that the blank serum had no significant effect on cell proliferation.After 48 h of treatment with 20%drug-containing serum,the cell shrinkage and rounding were observed under the microscope,and the number of cells decreased,indicating that the cell proliferation was significantly inhibited(p<0.05).After 24 h of treatment with 40%drug-containing serum,the cell shrinkage and rounding were observed under the microscope.The number of cells decreased,indicating that cell proliferation was significantly inhibited(p<0.05).Subsequently,40%drug-containing serum was selected to act on cells for 24 h.2.CCK8 results showed that AGS cells were observed as epithelioid adherent growth under the microscope,and the cells in the normal group grew well.Compared with the CK group,the cells in the blank serum group were observed under the microscope.There was no significant difference in each group,indicating that the blank serum had no significant effect on cell proliferation,and the drug-containing serum could be observed under the microscope.After the cells were treated,the cell contraction became more rounded and the number of cells decreased,indicating that the cell proliferation was significantly inhibited(p<0.05).3.RT-PCR results showed that compared with CK group,the expression levels of SLC7A11 and GPX4 in gastric cancer AGS cells were significantly decreased(p<0.05),and the expression level of P53 was significantly increased(p<0.05).4.Western Blot results showed that compared with CK group,the expression levels of SLC7A11 and GPX4 in gastric cancer AGS cells were significantly decreased,and the expression level of P53 was significantly increased(p<0.05).5.MDA results showed that compared with CK group,the serum containing AGS group could significantly reduce the expression level of MDA in gastric cancer AGS cells(p<0.05).6.GSH results showed that compared with CK group,the serum containing AGS group could significantly increase the expression level of GSH in gastric cancer AGS cells(p<0.05).Conclusion:Jiawei Qifang Weitong Granule can significantly inhibit the proliferation of gastric cancer cells,and its anti-tumor effect may be related to the inhibition of ferroptosis signal.
