| Background:Parkinson disease(PD)is a common neurodegenerative disorder characterized by motor slowness,tremors,rigidity,and postural instability.In addition to motor symptoms,psychiatric manifestations such as depressive symptoms are also common in PD patients.At present,the prevalence rate of depressive symptoms in PD has reached or even exceeded 50%,and only 20-25%of patients have been effectively treated.Depressive symptoms have become the most important factor that seriously affects the quality of life of PD patients and their caregivers.However,the pathogenesis of depressive symptoms in PD remains unclear.Studies have shown that genetic factors play a key role in the occurrence of depressive symptoms in PD.Several Parkinson disease-related genes(such as SNCA Repl gene,GBA gene,LRRK2019S gene)and depression disease-related genes(such as 5-HTTLPR gene)are associated with depressive symptoms in PD.Most of these studies are observational and lack rigorous clinical controls.A lot of 211 subjects were randomized controlled using a lot of propensity score method(PSM),and a lot of candidate genes are depression gene polymorphisms,norepinephrine gene polymorphisms,GABAergic system gene polymorphisms,and copper necrosis pathway gene polymorphisms.To explore the influence of above genes on the occurrence of depressive symptoms in PD patients.Methods:A total of 211 PD patients in Beijing hospitals were collected in this study.They were divided into depressed group and non-depressed group according to the Hamilton depression scale(HAMD)≥13 points,and 1:1 matching was performed according to the nearest neighbor principle.Factors such as sex,age,Disease course,drug dosage,Unified Parkinson Disease Rating Scale(UPDRS-Ⅲ)and Hoehn-Yahr Staging Scale were considered in matching,and the caliper value was 0.1.Finally,18 pairs of samples were included.The Agena platform was used to sequence 6 SNPS related to depression,7 SNPS related to GABAergic system,1 SNPS related to norepinephrine and 6 SNPS related to copper necrosis in the plasma of the 18 pairs of samples,and the differences in gene polymorphisms between the two groups were compared.SPSS 25.0 software was used for statistical analysis.Results:The comparisons between PD depressed group and non-depressed group was found is that(1)There were no significant differences of SNPs in depression-related genes,ncluding rs1432639,rs4143229,rs1554505,rs11643192,rs10149470 and rs62099069(P>0.05).(2)There was no significant difference of SNPS in GABAergic system,including rs211034,rs3219151,rs7725726,rs1992647,rs2256882,rs10036156,rs 13303344(P>0.05).(3)There were no significant differences of SNPrs2242446 in norepinephrine-related genes(P<0.05).(4)There were no significant differences of SNPs in cuproptosis related genes,including rs2233914,rs10488764,rs9535828,rs10981694,rs9535826 and rs1061472(P>0.05).Conclusion:The genetic basis of depressive symptoms in PD is different from that in common depression.Whether GABA,norepinephrine and cuproptosis related genes can be used as new drug treatment targets for PD depression and the basis of drug efficacy response selection remains to be further explored. |
