Objectives This study aimed to investigate the expression of coactivator-associated arginine methyltransferase 1(CARM1)and 54 k Da nuclear DNA and RNA-binding protein(p54nrb)in invasive breast cancer,and analyze their relationship with clinicopathological features in breast cancer.Further to investigate the correlation appeared between expression of CARM1 and p54 nrb in breast cancer and its clinical significance in breast cancer patients.Methods Retrospective clinical data from all breast cancer patients that were treated with modified radical mastectomy or radical mastectomy in the Department of Oncology of the affiliated Hospital of the North China University of Technology from January 1,2019,to July 31,2021,were collected.Cases were screened for relevance based on preset inclusion and exclusion criteria,121 cases were named as breast cancer group,and 30 cases of paracancerous normal tissues were randomly selected as a paracancerous group.Immunohistochemical SP method was used to detect the positive rate of CARM1 and p54 nrb expression in each group;Western blot method was used to detect the protein expression of CARM1 and p54 nrb in 3 pairs of fresh breast cancer tissues and their matched adjacent normal tissues.Moreover,we particularly investigated the correlation between CARM1 and p54nrb expression,as well as the correlation between their expression and clinicopathological paramenters of the tumors.Results 1 The positive expression rate of CARM1 protein in breast cancer group(55.4%)was significantly higher than that of paracancerous group(33.3%)(P<0.05).The positive expression rate of p54 nrb protein in breast cancer group(57.0%)was significantly higher compared with those in paracancerous group(30.0%)(P<0.05);2 The expression of CARM1 and p54 nrb were examined by Western blot.The result revealed that the expression level of CARM1 in the breast cancer group was higher compared with those in paracancerous tissues(P<0.05);The protein expression level of p54 nrb was significantly increased in the breast cancer group compared with the paracancerous group,showing a statistically significant difference(P<0.01);3 In breast cancer,the expression of CARM1 and p54 nrb were positively correlated with higher histological grade,ER&PR positive express and high Ki-67 index.However,the expression of CARM1 and p54 nrb were independent of age,tumor size,lymph node metastasis(N),TNM stage,and HER2 expression;4 The highest CARM1 expression rate was detected in HER2 overexpression breast cancer that was 70.8%,in Luminal B breast cancer was 67.9%,in triple negative breast cancer was 56.3%,and the lowest in Luminal A breast cancer was 41.5%.There was a significant difference in CARM1 expression among the four groups(P<0.05).The positive expression rate of p54 nrb was the highest in triple negative breast cancer tissues with a positive expression rate of 68.8%,and the lowest in Luminal A tissues,which was 49.1%.However,the difference test in the expression of p54 nrb between the four groups was not significant(P=0.365).5 The correlation between CARM1 and p54 nrb expression was further analyzed using Spearman rank correlation test,and the results showed that the expression of CARM1 was positively correlated with p54nrb(r=0.463,P=0.000).Conclusions 1 In breast cancer,elevated CARM1 and p54 nrb expression were significantly correlated with the occurrence and development of tumors.2 The high expression of CARM1 and p54 nrb was suggested that they may be related to the proliferation and high malignant degree of breast cancer.3 The expression of CARM1 is different in differently subtypes of breast cancer,which may play a supplementary role in the diagnosis,classification and treatment of breast cancer.4 A significant positive correlation between CARM1 and p54 nrb expression was detected in breast cancer.Figure 3;Table 8;Reference 129.
