The polysaccharides extracted from cultivated ginseng and adventitious root ginseng were named CGP and ARGP respectively.It was found that the molecular weight of ARGP was relatively small.In addition to Glc,the proportion of other monosaccharides in ARGP was higher than that in CGP.Through preliminary separation,it can be confirmed that compared with CGP,the proportion of acidic polysaccharides in ARGP is larger.In order to investigate the activity differences caused by different proportions,the polysaccharides after preliminary purification and mixing were named p CGP and p ARGP.Antioxidant activity and intestinal flora regulatory activity were compared.In terms of antioxidant activity,the free radical scavenging rates of p CGP and p ARGP at 1.5mg/m L were 15% and 50%,respectively.Polysaccharides extracted from different substances have different antioxidant activities.In addition,after removing small molecule interference,different polysaccharides were co cultured with intestinal flora.After co culture with p ARGP for 24 hours,the proportion of Lactobacillus increased sharply.The beneficial bacteria represented by Lactobacillus have broad-spectrum immunogenicity,which can improve the immune ability of human body.CGP-N(ARGP-N)and CGP-A2(ARGP-A2)were compared and studied.The infrared spectra,monosaccharide composition and molecular weight of ARGP-N(CGP-N)were similar.They’re all starch-like polysaccharides.The infrared spectra of CGP-A2 and ARGP-A2 showed the characteristics of ginseng pectin.The acidic polysaccharide obtained by elution with 0.3 M Na Cl was ginseng pectin.There are HG and RG-I domains in both kinds of ginseng pectins.It has been reported that pectin contains less than 75%galacturonic acid residues and has branching regions,especially the RG-I fragment of branching region,which plays an important role in stimulating immune response.The results showed that the contents of immune related bacteria were closely related to the proportion of neutral polysaccharide and acid pectin,and the proportion of different domains in ginseng pectin.In the preliminary exploration stage of ginseng gene editing,in order to regulate the editing target of ginseng polygalacturonate transferase,the corresponding sg RNA was designed and synthesized,and connected to the vector by homologous recombination to construct CRISPR/cas9 editing vector.Data indicated that CRISPR/Cas9 system of Pg_S1951.7 has positive effect to the accumulation of uronic acid in transgenic adventitious roots of P.ginseng.The culture conditions of adventitious roots of Astragalus membranaceus were optimized.When the concentration of IBA was 6 mg/L,the inoculation density was0.5%,and the medium p H was 5.5,the adventitious roots of A.membranaceus grew best.The results showed that the contents of astragaloside Ⅳ and calycosin-7-O-β-D-glucoside in four kinds of roots were up to the Chinese Pharmacopoeia 2020.In the application of elicitor,the content of astragaloside IV reached 0.9% after applying 50 mg / L penicillium sp.crude extract for 7 days,which was 2.5 times of the control group.
