【Objective】 To study the changes of the expression levels of calcium failure protein(CSEN),peripheral protein(Prph)and ApoJ in human serum after diffuse axonal injury,and explore the application value of biomarkers CSEN,Prph and ApoJ in forensic medicine diagnosis.【Methods】 Enzyme-linked immunosorbent assay(ELISA)was performed on CSEN,Prph and ApoJ of all serum samples.All serum samples were set with multiple Wells,and CSEN,Prph and ApoJ were quantitatively detected respectively,and the measured concentrations were averaged.Graph Pad Prism 9 software was used to conduct T test and variance analysis for the concentration of the sample,and the graph was drawn.【Results】(1)Compared with the control group,CSEN,Prph and ApoJ were all highly expressed in the brain injury group,and there were significant statistical differences among them(P<0.0001).(2)Compared with the control group,the expression levels of SERUM Prph and ApoJ in the brain injury group were statistically different in the young,middle-aged and elderly groups;The expression level of serum CSEN in the brain injury group was only statistically different in the middle-aged group,and there was no statistical difference between the young group and the elderly group.(3)There were no significant differences in serum CSEN,Prph,ApoJ,expression levels in different stages of brain injury compared with acute stage(≤7d)and chronic stage(>7d).(4)There was no statistical difference in the expression levels of CSEN,Prph and ApoJ in the mild,moderate and severe hemolysis samples between the control group and the brain injury group.(5)The expression levels of CSEN,Prph and ApoJ in serum were increased by different causes of TBI,and there was no statistical difference in the degree of effect.(6)The expressions of Prph and ApoJ in serum of normal people were not affected by different causes of death,but CSEN in serum of normal people were affected by sudden cardiac death and septic shock.【Conclusion】 This experiment preliminarily verified the feasibility of serum CSEN,Prph and ApoJ as biomarkers of diffuse axonal injury.Serum CSEN,Prph and ApoJ were highly expressed after diffuse axonal injury.The expression levels of CSEN,Prph and ApoJ in serum were not affected by different injury process,hemolysis degree of different serum samples and different causes of injury,but the expression levels of CSEN,Prph and ApoJ were stable.In addition,the expression levels of serum Prph and ApoJ increased in different age groups after TBI,while the expression level of serum CSEN increased only in the middle-aged group.These results suggest that Prph and ApoJ have greater potential as biomarkers for DAI than CSEN.
