Camellia oleifera Able,belonged to genus Camellia Linn(Theaceae),is one of the pure natural high-grade oil plants unique to China.Different parts of the titled plant such as leaves,seeds,roots,fruit shells and seed meal containing active components,are of a variety of medicinal uses,and bear high medicinal value.However,the research on the chemical components and pharmacological activities of Camellia oleifera husk is very limited.Therefore,in order to enrich the chemical components in the husk of Camellia oleifera,and provide reference basis for further discovering its bioactive compounds and pharmacological research,this thesis mainly separates and identifies the chemical components of the Camellia oleifera husk.In this study,95% ethanol extract from Camellia oleifera husk was used to isolated and purified components by using open silica gel,gel column,ODS column chromatography and semipreparative liquid chromatography.UV,IR,MS,NMR and other spectroscopic methods were used to determine or identify the structures of compounds.34 compounds were isolated and identified.Of them,two compounds were new compounds.The 34 compounds are as follows:(-)-isolariciresinol-9’-methyl succinate(CC-1),(-)-isolariciresinol-9-methyl succinate(CC-2),(+)-epipinoresinol(CC-3),(1S,2R,5S,6R)-2-(4-hydroxyphenyl)-6-(3-methoxy-4-hydroxyphenyl)-3,7-dioxabicyclo[3.3.0]octane(CC-4),syringaresinol(CC-5),(+)-pinoresinol(CC-6),salicifoliol(CC-7),(-)-secoisolariciresinol(CC-8),arctigenin(CC-9),(+)-isolariciresinol(CC-10),pinnatifidanin B VI(CC-11),pinnatifidanin B V(CC-12),dihydrodehydrodiconiferyl alcohol(CC-13),1-(4-hydroxyphenyl)ethanone(CC-14),3-hydroxy-4-methoxybenzoic acid(CC-15),protocatechuic acid methyl ester(CC-16),trans-2-hydroxy cinnamic acid(CC-17),3-hydroxy-4,5-dimethoxyb enzoic acid(CC-18),4-hydroxy benzaldehyde(CC-19),veratroylglycol(CC-20),2,3-dihydroxy-1-(4-hydroxy-3,5-dimethoxyphenyl)-1-propanone(CC-21),1-(3’,5’-dimethoxy)phenyl-2-(4’’-hydroxy)phenylethane(CC-22),loliolide(CC-23),3-isopropyl-5-acetoxycyclohexene-2-one-1(CC-24),(6R,7E,9R)-9-hydroxy-4,7-megastigmadien-3-one(CC-25),(+)-dehydrovomifoliol(CC-26),7-epiloliolide(CC-27),5,6,7,8,3’,4’-hexamethoxyflavone(CC-28),3,5,6,7,8,3’,4’-heptamethoxy-flavone(CC-29),taxifolin(CC-30),naringenin(CC-31),(3R,9R,10R)-panaxytriol(CC-32),aurantiamide(CC-33),1H-indole-3-carbaldehyde(CC-34).CC-1 and CC-2were new lignans.Compounds CC-3~CC-4 and CC-7~CC-34 were isolated from Camellia oleifera for the first time.The antitumor activity and antioxidant activity of all the compounds were evaluated,and it was found that except CC-32,which had weak cytotoxic activity against A375(human malignant melanoma cells)and Huh-7(human hepatoma cells),other compounds had no effect on Huh-7(human hepatoma cells),H460(human lung cancer cells),MCF-7(human breast cancer cells),A375(human malignant melanoma cells)and HCT116(human colon cancer cells).The antioxidant activity of all compounds was determined by free radical scavenging(DPPH)experiment,and the results showed that CC-1,CC-2,CC-5,CC-6,CC-30 had weak antioxidant capacity against DPPH,while CC-3,CC-8,CC-10,CC-16 have good antioxidant capacity. |
