Objective:To explore the role of polyadenosine diphosphate ribose polymerase-1 in myocardial ischemia and reperfusion injury in Sanguis Draconis flavones(SDF),and to explore the potential mechanism of total flavonoids on myocardial ischemia and reperfusion injury.Methods:The 40 clean-grade rats fed for 14 days were equally divided into four groups according to numerical randomization: blank control group(Contorl group),sham group(Sham group),myocardial ischemia and reperfusion group(group I / R),and dragon blood depletion total flavonoids treated group(SDF group).Each group contained 10 rats.Equal amount of normal saline,blank control group: equal amount of normal saline on time without mold creation;sham group: no ligation below the left anterior descending branch;myocardial ischemia-reperfusion group: tie the anterior descending artery of the left coronary artery in rats for 30 min and then cut the ligation line for 2h to construct a model of myocardial ischemia-reperfusion injury;total flavonoids treatment group: gastric SDF mixture(180 mg /kg)once for 14 consecutive days before mold creation,and the subsequent molding procedure was the same before.ECG was recorded at three time points before ischemia,30 min after ischemia and 2h reperfusion,Rat myocardial tissue and abdominal aorta serum samples were obtained immediately after the establishment of each rat model,Chlorinated triphenyl tetrazolium(Triphenyl Tetrazoliumchloride,TTC)stained the infarct area of rat myocardium,hematoxylin-Eosin(Hematoxylin-Eosin,HE)staining to observe the cellular structure and pathological changes of myocardial tissue;Rat serum lactate dehydrogenase(Lactate Dehydrogenase,LDH)and creatine kinase isoenzymes(Creatine Kinase Isoenzyme,CK-MB)in the concentration.The mRNA expression level of poly SYDPK ribose polymerase-1(Poly ADP-ribose Polymerase1,PARP-1)in rat myocardial tissue was determined by q-PCR.Results:(1)Serum concentrations of CK-MB and LDH in Sham group were not significantly different compared with Control group,and TTC staining results indicated that there was no statistical difference in myocardial infarction area(P>0.05);serum concentrations of CK-MB and LDH increased significantly compared with Sham group,and myocardial infarction area significantly increased(P<0.05);serum CK-MB and LDH in SPF rats decreased significantly compared with I/R group(P<0.05).(2)Under microscopic observation of HE slices,the nuclei in Control group and Sham group were medium and clear,the morphology was complete,the myocardial collagen fibers were orderly,and the myocardial cell structure was clear and neat,and no inflammatory cell infiltration was seen.In the I/R group,massive cardiomyocyte degeneration and necrosis,edema between the muscles,uneven staining,disordered arrangement of myocardial collagen fibers,and massive infiltration of inflammatory cells were observed.The SDF treatment reduced the pathological damage of cardiomyocytes.(3)The expression of PARP-1 mRNA in Sham group compared with Control(P>0.05);the expression of PARP-1 mRNA in I/R group compared with Sham group(P<0.05),the expression of PARP-1 mRNA decreased compared with I/R group(P<0.05),and SDF may reduce myocardial reperfusion injury by downregulating PARP-1mRNA expression in cardiomyocytes..Conclusion:(1)Total flavonoids can protect cardiomyocytes and thus reduce myocardial ischemia and reperfusion injury in rats.(2)Total flavonoids may protect the cardiomyocytes of MIRI rats by downregulating the expression of PARP-1 mRNA in myocardial tissue.
