Objective: To explore the key target genes closely related to the metastasis and survival prognosis of Anaplastic thyroid cancer,and to supplement the molecular markers of Anaplastic thyroid cancer.Methods: Firstly,the clinicopathological data of Anaplastic thyroid cancer were collected,and the correlation between survival and prognosis was analyzed based onthe patient’s age,gender,TNMstage,lymph nodes,and distant metastasis.Secondly,we selected the GSE53072 gene chip data from the GEO public database,identified the gene target genes and Hub genes with significant differential expression(P<0.01)in Anaplastic thyroid cancer through the online database of Network Analyst and the Cytoscape software,and used We Gestalt and KOBAS databases to perform GO enrichment analysis and KEGG pathway analysis on the target genes and Hub genes to screen out the key target genes of Anaplastic thyroid cancer,Using STRING database,protein interaction network analysis was performed on key target genes.Finally,download the clinical data of Anaplastic thyroid cancer including TP53 and TERT key target genes in the c Bio Portal database,and analyze and verify the correlation between TP53 and TERT key target genes and age,gender,tumor stage,lymph node and distant metastasis,survival and prognosis in Anaplastic thyroid cancer.Results:(1)In 33 patients with Anaplastic thyroid cancer,lymph node metastasis and distant metastasis were significantly correlated with survival and prognosis(P<0.01).(2)Based on the GEO database,a total of 2797 differentially expressed genes were selected from the GSE53072 dataset(specimens: Anaplastic thyroid cancer and normal tissues),including1092 highly expressed genes and 1705 low expressed genes(corrected p<0.05 and absolute log FC ≥ 1).(3)Finally,13 key target genes were selected: TP53,TERT,PLK1,BUB1,MAD2L1,PTEN,CDC20,UBE2 C,TPO,TG,IYD,FN1,COL1A1.Among them,the key target genes closely related to lymph node metastasis,distant metastasis,and survival and prognosis of Anaplastic thyroid cancer are TP53,TERT.(4)Key target genes in Anaplastic thyroid cancer can participate in biological regulation,thyroid hormone synthesis,cell cycle,cell adhesion molecules,regulation of tumor immune infiltration levels,and PI3K-Akt signaling pathways.(5)In Anaplastic thyroid cancer,there were significant differences in age,lymph node metastasis,distant metastasis,and survival and prognosis between the TP53 and TERT gene mutation groups and the non mutation group(P<0.05).(6)TP53 was positively correlated with B lymphocytes,CD4+T cells,macrophages,neutrophils,and dendritic cells in tumor immune infiltration(r>0 and P<0.05 for all);TERT was positively correlated with B lymphocytes,CD8+T cells,macrophages,neutrophils,and dendritic cells(r>0 and P<0.05 for all).Conclusion:(1)The survival and prognosis of Anaplastic thyroid cancer has been proven to be closely related to lymph node metastasis and distant metastasis.(2)Explore 13 key target genes for Anaplastic thyroid cancer,including TP53 and TERT,which are closely related to lymph node metastasis,distant metastasis,and survival and prognosis of Anaplastic thyroid cancer.(3)TP53 and TERT are closely related to the metastasis and survival prognosis of Anaplastic thyroid cancer,and are expected to become potential molecular markers for the study of Anaplastic thyroid cancer,expanding new research directions for the study of Anaplastic thyroid cancer.
