Effect Of Dendrobium Candidum On Endoplasmic Reticulum Stress-Related Proteins In Renal Tissue Of Chronic Glomerulonephritis Rats

Objective:To investigate the effect of Dendrobium Candidum(DC)on Endoplasmic reticulum stress(ERS)in the kidney of rats with Chronic Glomerulonephritis(CGN)and the therapeutic effect of DC on the Glomerulonephritis of CGN.Methods:(1)Forty Sprague-dawley rats were randomly divided into model group(N= 30)and normal group(N=10).Chronic glomerulonephritis models were established by intraperitoneal injection of bovine serum albumin acid(BSA),20 rats of model group were randomly divided into two groups,10 rats in each group were treated with different doses of Dendrobium officinale dry powder.(2)The rats in the normal group and the rest model group were given the same volume of normal saline by Gavage,and all groups were continuously gavaged for 8 weeks.(3)The general condition,24 h urinary protein,creatinine and urea nitrogen were observed.(4)The pathological changes of renal tissues were observed and compared under light microscope.The expression of GRP78,p-PERK and CHOP proteins in endoplasmic reticulum were detected by immunohistochemistry.Results:(1)Dendrobium officinale can effectively improve the general condition of chronic glomerulonephritis rats.After 8 weeks of oral administration of Dendrobium officinale,the state of the rats in the treatment group was better than before,their activities were more sensitive than before,and their drinking water and eating were increased than before,the hair was shiny,but its general condition was not as good as that of the normal rats.(2)After modeling,the 24-hour urine protein of CGN rats in each group was significantly higher than that before modeling(P<0.01).After intervention of Dendrobium candidum,the24-hour urine protein of treatment and high-dose groups were significantly lower than that after modeling(P<0.01),but there was no difference between the two groups(P>0.05).(3)After treatment with Dendrobium candidum,the serum creatinine and blood urea nitrogen of CGN rats in each group decreased(P<0.01),and the renal function was improved to some extent,but there was no difference between the two treatment groups(P<0.05).(4)Compared with the normal group,mesangial cell proliferation,widening of mesangial area,thickening of basement membrane,Telangiectasia hyperemia of glomerulus,dilatation of glomerular saccule,thickening of saccule wall,and large amount of glycogen deposition were observed in the model group,the area percentage of renal interstitial fibrosis was significantly increased(P<0.01),which was improved to some extent by Dendrobium candidum intervention(P<0.01),and the improvement effect of high dose group was more obvious than that of treatment dose group(P<0.05).(5)After the intervention of Dendrobium candidum,the expression of endoplasmic reticulum stress-related proteins GRP78,p-PERK and CHOP in the renal tissue of CGN rats decreased compared with the model group,and their optical density decreased compared with the model group(P<0.05 or P<0.01).Conclusion:(1)Dendrobium officinale can improve 24-hour urinary protein excretion,improve blood biochemical indexes,and reduce the production of toxins such as creatinine and urea nitrogen in chronic glomerulonephritis rats.(2)Dendrobium candidum can reduce the proliferation of mesangial cells and mesangial matrix,relieve renal fibrosis and renal interstitial edema.(3)Dendrobium candidum may inhibit the development of endoplasmic reticulum stress by reducing the production of GRP78,p-PERK,CHOP and other endoplasmic reticulum stress-related proteins.
