| Objective: To explore the regenerative medical technology(Moist Exposed Burn Therapy/Moist Exposed Burn Ointment,MEBT/MEBO)by treating the stromal cell-derived factor-1in rats(SDF-1),chemokine receptor 4(CXCR4)expression level effects to accelerate the repair process of chronic wound-healing.Methods:(1)SPF male Wistar rats 90 according to random number table method was randomly divided into blank group(KBZ group),acute group(JXZ group),model group(MXZ group),recombinant bovine basic Fibroblast Growth Factor group(BFZ group)and MEBO group(MBZ group),each group 18,blank group skin shaving without other skin injury treatment,acute group do whole skin treatment,establish acute wound,model group,MEBO group and BFZ group refer to full layer skin defect method and Shen’s modified plastic ring granuloma quantitative method to establish chronic difficult healing wound model.(2)Wound surface treatment after successful mold making:JXZ and MXZ groups were covered with saline gauze,BFZ wounds were treated with new drugs,and MBZ wounds with MEBT / MEBO.Before dressing change,1/5000 furacillin solution was disinfected,and the dressing frequency was twice per day.(3)After the preparation of the animal model,6 rats were randomly selected from each group at 3d,7d and 14 d,and the whole wound granulation tissue was cut after anesthesia.Part of the specimen was fixed in formaldehyde,then stained with HE staining and Masson staining.The remaining wound tissues were placed in-80℃refrigerator,which was used to detect SDF-1 and CXCR4 m RNA expression levels and protein expression levels in the wound tissues by Real-time q PCR and Western Blot.Results:(1)Observation of wound healing: on the 3rd day of mold making,no significant healing in the four groups,a small amount of fresh granulation tissue in JXZ group,and no obvious granulation tissue in MXZ,MBZ and BFZ groups,among which the wounds in MXZ group had more exudation and the edema was obvious.On the 7th day of mold making,except for the MXZ group,the other three groups were reduced,while the JXZ group was bright red,no ooze edema,the granulation of the MBZ and BFZ groups was fresh than before,no exudation,and the MXZ group had little exudation and edema,and necrotic tissue was visible.On the 14 th day of mold making,the wound of JXZ group was basically healed,some MBZ and BFZ groups were close to healing,the wound granulation tissue was fresh,and the wound epithelialization was obvious.In the MXZ group,the wound surface had small exudation and edema,and necrotic tissue was visible.rb-b FGF and MEBT/MEBO are effective in treating chronic difficult-healing wounds,and the efficacy is not significantly different.(3)Protein expression of SDF-1 and CXCR4 and m RNA transcript levels in the wound tissues of rats:compared with the KBZ group,on the 3rd,7th,and 14 th days after mold modeling,the protein expression of SDF-1 and CXCR 4 of JXZ,BFZ and MBZ increased,and were statistically different(P <0.05).In the MXZ group,the expression was significantly higher on days 7 and 14.On day 14,the expression level of the MXZ group was higher than that of the other four groups(P <0.05).At this time,the JXZ wound is near the healing state.Conclusion:(1)From the results from this study,MEBT/MEBO can shorten the inflammatory period of chronic difficult healing wounds and accelerate the healing process of chronic wounds.(2)Treatment of MEBT/MEBO may promote chronic refractory wound healing by affecting SDF-1,CXCR4 protein expression levels and m RNA transcript levels. |
