| Objective By studying the effects of Yiqi wenyang jianpi prescription on colonic VIP,5-HT and SP in rats with laxative constipation,the possible mechanism of its treatment for laxative constipation was discussed.Methods:Fifty SD rats with a body weight of 180-220 g were selected,half male and half female,and housed in cages with different genders.They were randomly divided into blank group(10),model group(10),Moxapride group(10),Yiwenyangjianpi prescription low-dose group(10)and Yiwenyangjianpi prescription high-dose group(10).The blank group was fed with 10ml/kgd distilled water and the other four groups were fed with ordinary dry feed and 10ml/kgd rhubarb no-decoction water solution.The first dose was200mg/kg·d,followed by an increasing dose of 200mg/kg·d,maintained when more than half of the rats had loose stools,and continued to increase at 200mg/kg·d when loose stools disappeared,with four cycles and the final rhubarb dose of 3200mg/kg·d for 3 months.After withdrawal of the drug,ordinary feed was fed to be treated.One week later,the drug intervention group was divided into groups.After 4 weeks of continuous administration,the number of fecal granules in each group for 24 hours was observed and recorded.Intestinal transport function was detected in each group.The contents of VIP,5-HT and SP in colon tissues were detected.Results(1)The number of stool at 24h:(1)The amount of stool at 24 h in the drug intervention group was significantly higher than that in the modeling group at the 4th week(P<0.05).(2)Compared with the first week of treatment,24 h stool volume in the western medicine group and the Chinese medicine group was significantly increased at week 4(P<0.05).(3)Compared with Western medicine group,stool volume of Yiqen-Yang-Jianpi prescription low-dose group was significantly increased at 24 h after drug treatment(P<0.05).(2)Colon transmission function:(1)Compared with blank group,the colon transmission function of the model group was significantly decreased(P<0.05).(2)Compared with the modeling group,the transmission function of colon in the drug intervention group was significantly increased(P<0.05).(3)Compared with the western medicine group,the colon transmission function of rats in Yiwenyang Jianpi formula high-dose group was significantly increased(P<0.05).(3)VIP:(1)Compared with blank group,the content of VIP in colon of the model group was significantly increased(P<0.05).(2)Compared with the modeling group,VIP of colon in the drug intervention group was significantly decreased(P<0.05).(3)Compared with Western medicine group,the decrease of Yiwenyang Jianpi formula high-dose group was more obvious(P<0.05).(4)5-HT:(1)The 5-HT levels in the modeling group were significantly lower compared with the blank group(P<0.05).(2)Compared with the model group,the 5-HT levels in the colonic tissues of rats in the treatment group were significantly higher(P<0.05).(3))Compared with the western medicine group,the 5-HT in the colon tissue of rats in the high dose of Chinese medicine was more significantly elevated(P<0.05).(5)SP:(1)Compared with blank group,SP content was significantly decreased(P<0.05).(2)Compared with model group,SP content in colon of rats in drug intervention group was significantly increased(P<0.05).(3)Compared with western medicine group,SP of colon tissue in Yiwenyang Jianpi formula high-dose group was increased more significantly(P<0.05).Conclusion1.Yiqi wenyang jianpi prescription can Improving intestinal transmission function in rats with laxative constipation and increasing the number of stools in 24 h.2.Yiqi wenyang jianpi prescription can reduce the content of colonic VIP in rats with laxative constipation,so as to regulate gastrointestinal movement.3.Yiqi wenyang jianpi prescription can reverse the change of 5-HT level in colon of rats with laxative constipation,increase the content of 5-HT in colon tissue of rats,so as to regulate gastrointestinal motility,promote intestinal peristalsis and improve constipation symptoms.4.Yiqi wenyang jianpi prescription can reverse the change of SP level in colon of rats with laxative constipation,increase the content of SP in colon tissue of rats,so as to regulate gastrointestinal motility,promote intestinal peristalsis and improve constipation symptoms. |
