Objective:To provide a theoretical and experimental basis for the clinical application of the homology of medicine and food," Huangjing"(Polygonati Rhizome),and to provide reference and guidance for the exploration of dietary interventions for the effective prevention and treatment of diabetic cardiomyopathy through in-depth excavation of the ancient literature of "Huangjing" and bibliometric analysis of modern literature features and research trends,as well as to verify the effect mechanism of Polygonatum sibiricum polysaccharide against diabetic cardiomyopathy through experimental studies.Methods:1.Ancient literature mining: we searched and collected all the articles related to "Huangjing" in the Chinese Medical Dictionary(5th edition),and applied Excel 2019 to statistically analyze the efficacy,ancient prescriptions and drug combinations of "Huangjing" in different periods.2.Bibliometric analysis of modern literature: based on Cite Space visualization software,we analyzed the research status and development trend of "Huangjing".3.Experimental study: The rat model of type 2 diabetes mellitus was replicated by high-fat diet combined with streptozotocin,and two dose groups of 200 mg and 400mg/kg were intervened continuously for 8 weeks,through observing the effects of Polygonatum sibiricum polysaccharide on general signs,blood glucose,myocardial histomorphology(HE and Masson staining)of the model rats.And investigated the mechanism of Polygonatum sibiricum polysaccharide on the prevention and treatment of diabetic myocardial injury by using Western blot,RT-q PCR and other experimental methods and techniques.Results:1.Ancient literature excavation: a total of 156 ancient texts and 356 articles of "Huangjing" were included.It contains 12 categories of ancient texts,which have been recorded in relevant ancient texts in all generations since Han Dynasty.In addition to the categories of materia medica,medical scriptures,prescription books,medical cases,etc.,there are also more records in the monographs of health and food therapy.The efficacy of "Huangjing" is slightly different in the ancient herbal texts,but all emphasize its tonic effect and food value.A total of 56 formulas containing "Huangjing" were compiled,and126 Chinese herbal medicines with "Huangjing" were counted,among which the more frequent ones were Chinese wolfberry,ginseng and other medicinal and dietary tonic herbs.The results showed that the combination of "Huangjing" with various herbs mainly played a role in improving the effects of asthenic fatigue injury and internal heat dispersion-thirst.2.Bibliometric analysis of modern literature: a total of 2385 papers were included,with an overall upward trend in the number of publications.The institutions to which the high-yielding authors belonged were mostly Chinese medicine universities in various provinces.The authors’ knowledge map showed that the cooperation team formed in China was not very tightly cooperated,and the three cooperative teams formed abroad were more closely cooperated.The keyword co-occurrence and clustering results showed that Polygonatum sibiricum polysaccharide,antioxidant,TCM treatment,and diabetes were the hot spots of research in this field.The results of keyword co-occurrence show that process of "Huangjing" and experimental research are the research frontiers in this field.3.Experimental studies:(1)Compared with the normal group,the rats in the model group were depressed,lacked luster of the fur,and had significantly increased diet,water consumption and urine output.(2)Compared with the normal group,after 8 weeks,the blood glucose and glycosylated hemoglobin levels were significantly higher(P<0.01)and insulin levels were significantly lower(P<0.01)in the model group rats;the echocardiographic test results showed that the ejection fraction and shortening fraction values were significantly lower(P<0.01)in the model group rats;the HE and Masson results suggested that the model group rats Myocardial tissue showed significant pathological changes such as fracture,fibrosis and inflammatory cell infiltration;myocardial tissue CK,BNP,c TNI and MDA levels were significantly increased(P<0.01)and GSH and SOD were significantly decreased(P<0.01)in the model group rats.(2)Compared with the model group,blood glucose and glycated hemoglobin were significantly lower(P<0.05 and P<0.01)and INS levels were significantly higher(P<0.01)in the model rats after intervention in each dose group of Flavopiridium polysaccharide;EF and FS values were significantly higher(P<0.05)in rats;HE and Masson results also suggested that myocardial histopathological damage was significantly improved in the model rats;myocardial tissue of rats CK,BNP,c TNI and MDA levels were significantly decreased(P<0.01),and GSH and SOD were significantly increased(P<0.01).(3)The protein and m RNA expressions of NLRP3,Caspase-1 and GSDMD in myocardial tissue of rats in the model group were significantly increased compared with the normal group(P<0.01).(4)The protein and m RNA expressions of NLRP3,Caspase-1 and GSDMD in myocardial tissue were significantly decreased in each dose group of Polygonatum sibiricum polysaccharide compared with the model group(P<0.01).Conclusion: "Huangjing" has been used as a medicinal and dietary supplement by medical practitioners since ancient times,with the effects of Nourishing yin and moistening lung,tonifying spleen and kidney and is also often combined with other medicinal and dietary substances to form tonic formulas.The modern research of "Huangjing" as a whole shows a growing trend,and the current research hotspots are Polygonatum sibiricum polysaccharide,antioxidant,diabetes,etc.The research trend gradually evolves to the fields of Flos Lobata concoction,experimental research,etc.There is a problem of low cooperation among authors,institutions and countries.The Polygonatum sibiricum polysaccharide can inhibit inflammatory response and oxidative stress to protect cardiomyocytes,significantly improve diabetes-induced morphological,structural and functional alterations of the heart,and has a significant role in preventing and treating diabetic cardiomyopathy;over-activation of NLRP3/Caspase-1/GSDMD signaling pathway may be an important molecular mechanism of diabetes-induced myocardial injury.
