| Background To concentrate on the differential metabolites in serum of progressive vitiligo patients and healthy controls.Objective Use HPLC/MS to analyze the characteristics of serum metabolites between advanced vitiligo and healthy controls,to screen different metabolites and to find out the differences of serum metabolites and characteristic compounds between patients with advanced vitiligo and healthy controls,so as to provide a new perspective for revealing the pathogenesis of vitiligo and clinical diagnosis and treatment of vitiligo.Method(1)sample collection and grouping: from October 2020 to June 2021,patients with vitiligo were recruited in the Department of Dermatology,the first affiliated Hospital of Anhui Medical University.The diagnosis was based on skin depigmentation spot and Wood lamp examination.Two dermatologists diagnosed and evaluated the patients at the same time.The healthy control volunteers came from the physical examination center of our hospital.After obtaining the consent of the patients and their guardians,they all signed the informed consent to participate in the vitiligo study,and then collected the clinical data and fasting peripheral venous blood samples of all volunteers.The study was approved by the Ethics Committee of Anhui Medical University.(2)Using high performance liquid chromatography-mass spectrometry technology to comprehensively scan and detect the extracted qualified samples,combined with metabonomics analysis method to explore the characteristics of serum metabonomics of patients and volunteers.The original mass spectrometry data were imported into Thermo Scientific "Compound Discoverer" software,and the mass spectrometry data obtained in this study were processed.After SERRF correction,the original mass spectrometry data were imported into the corresponding Metaboannlyst website for qualitative and quantitative analysis.Finally,the differential metabolites were screened by Simca14.1 software,multivariate statistical analysis and univariate statistics,and the data were visualized by R language.Through statistical processing,we looked for the differential metabolites between the sera of patients with progressive vitiligo and healthy people,and then found the differential metabolic pathways associated with progressive vitiligo through KEGG database.Results(1)In the PCA unsupervised model analysis,the quality control aggregation was good,but there was no significant difference between the progressive vitiligo group and the healthy control group,while in the OPLS-DA model analysis,the separation effect between the two groups was good,indicating that there was a significant difference between the two groups,indicating that there were some differences in serum metabonomic characteristics between the two groups;(2)A total of 336 metabolites were detected in serum samples from patients with progressive vitiligo and healthy controls.there were 10 different metabolites between the two groups,including downregulated metabolites: piperine;pantothenic acid;up-regulated metabolites:1-Stearoylglycero,Stearoylethanolamide,Imidazoleacetic acid,Stearic acid,cis-5,8,11,14,17-Eicosapentaenoic acid,Oleoyl ethanolamide,Bilirubin,Uridine.We used Metabo Analyst 5.0 platform and KEGG database to analyze the differential metabolites.The identified 10 differential metabolites can map 8 metabolic pathways: Biosynthesis of unsaturated fatty acids;pantothenic acid and Co A biosynthesis pathway;alanine metabolic pathway;vitamin digestion and absorption pathway;histidine metabolic pathway;fatty acid biosynthesis pathway,pyrimidine metabolism pathway and porphyrin and chlorophyll metabolism pathway.Among them,the biosynthesis metabolism pathway of unsaturated fatty acids was significantly different between the two groups.Conclusion There are significant differences in serum metabolism between patients with progressive vitiligo and healthy controls.Combined with 10 related metabolic biomarkers,significant metabolic pathway of biosynthesis of unsaturated fatty acids were excavated.It can provide a powerful scientific basis for further exploring the pathogenesis of vitiligo and provide a new perspective for clinical diagnosis,treatment and prognosis of vitiligo. |
