Prostate cancer ranks second in the incidence of malignant tumors in men,and its proportion is gradually increasing.Prostate cancer is not easy to detect because the early clinical symptoms are not obvious or absent,and most of the patients are found in the middle and advanced stages,so early detection and early treatment are very important for the diagnosis and treatment of prostate cancer.Drugs for prostate cancer,such as abiraterone and enzalutamide,often have poor treatment results because of tumor resistance,so it is necessary to actively seek new therapeutic targets.Tandem C2 domain nucleoprotein(TC2N)is a protein containing the C2 domain and belongs to the carboxyl-terminal(C-type)tandem C2 protein family.Studies have shown that TC2 N plays the role of oncogene or tumor suppressor gene in cancer,but its role in prostate cancer has not been explored.This study intends to explore the role and molecular mechanism of TC2 N in the progression of prostate cancer.Obective: To investigate the role of tandem C2 domain nuclear protein(TC2N)in prostate cancer and its molecular mechanism.Methods: We first used GEO and GEPIA online tools to analyze the differences in TC2 N expression in tumors;Western Blotting and immunohistochemistry were used simultaneously to analyze TC2 N expression in prostate cancer samples and their normal prostate tissues.Then,Linked Omics and Metascape tools were used to analyze TC2 N co-expression related genes and their GO and KEGG pathways.The String database was used to analyze the relationship between TC2 N and its protein interaction.Survival analysis using the GEPIA online tool.We subsequently determined TC2 N expression in prostate cancer cell lines using Western Blotting by knocking down the TC2 N gene in PC3 and DU145 cells;Through cell scratch experiment,clone formation experiment,Transwell invasion experiment,CCK-8 and EDU cell enhancement experiment,its effect on the growth process of prostate cancer cell line was explored.Finally,in terms of mechanism research,the GEPIA online tool was used to analyze the correlation between transcription factor 3(STAT3)and TC2 N.Use GO BP and GO CC data to analyze the functional connection between the two;Western Blotting experiments were used to study its effect on the expression of transcription factor 3(STAT3)and its phosphorylated protein.Results: TC2 N expression in prostate cancer tissues is higher than in paracancerous tissues(p<0.05),and patients with high expression levels have lower overall survival.GO analysis found that TC2 N and its related genes are mainly involved in the proteolytic function of cell transport and cellular protein hydrolysis.KEGG pathway analysis showed that TC2 N was closely related to protein processing,reticulation,m RNA monitoring pathway,and T cell receptor signaling pathway in the endoplasmic reticulum.Regulation of T cell activation and adaptive immune responses.The results of protein interaction network showed that TC2 N had a strong co-expression relationship with MMRN1,CNGB1,SMEK1,SCARA5,CATSPER2,CLEC4 M,STXBP5,CATSPERB,OTUD3 RASAL2 and other proteins.The results of cell scratch experiment,clone formation experiment,Transwell invasion experiment,CCK-8 and EDU cell value-added experiment showed that knockdown TC2 N could inhibit the migration,invasion and proliferation of prostate cancer cell lines.The GEPIA online tool analyzed that TC2 N and STAT3 expression were positively correlated,and Western Blotting test results showed that knocking down TC2 N could reduce STAT3 and its phosphorylation expression,indicating that STAT3 was involved in TC2 N regulation.Conclusion: This study shows that tandem C2 domain nucleoprotein(TC2N) expression is upregulated in prostate cancer,and its high expression indicates a poor prognosis.At the same time,the expression level of TC2 N was positively correlated with the migration,invasion and proliferation capacity of prostate cancer cell lines.In terms of mechanism,TC2 N regulates the migration,invasion and proliferation of prostate cancer cell lines through the STAT3 pathway under in vitro culture conditions,revealing that TC2 N may be a potential biomarker and therapeutic target for prostate cancer patients.
