Objective:The objective of this study was to investigated the effects of ellagic acid from pomegranate peel extract on the main periodontal pathogenic bacteria Porphyromonas gingivalis and Fusobacterium nucleatum,including the effects on bacterial and biofilm growth and adhesion,through in vitro tests.The study aims to investigate the mechanism of ellagic acid on periodontal disease control.Methods:1.Ellagic acid monomer was dissolved in dimethyl sulfoxide(DMSO)to determine the minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of ellagic acid against Porphyromonas gingivalis and Fusobacterium nucleatum by liquid dilution method.2.The bacterial adhesion ability and growth curve were measured by ULTRAVIOLET spectrophotometer.3.The minimum biofilm inhibition concentration(MBIC50)and the minimum biofilm reduction concentration(MBRC50)were determined by biofilm crystalline violet staining.Result:1.The MICs of ellagic acid against Porphyromonas gingivalis and Fusobacterium nucleatum were 1g/L and 1g/L,and the MBCs were 2g/L and 2g/L,respectively.2.The inhibition of bacterial adhesion of Porphyromonas gingivalis and Fusobacterium nucleatum,at 1 g/L was 80.68%and 83.53%(p<0.05);the effect of ellagic acid on the growth of Porphyromonas gingivalis and Fusobacterium nucleatum,was to delay the logarithmic growth period of Porphyromonas gingivalis from 8 h to 12 h and that of Fusobacterium nucleatum,from 4 h to 12 h.3.The MBIC50 and MBRC50 of ellagic acid against Porphyromonas gingivalis and Fusobacterium nucleatum were 4 g/L and 8 g/L.Conclusions:Ellagic acid was able to inhibit the growth and adhesion of Porphyromonas gingivalis and Fusobacterium nucleatum,showing effective inhibition and biofilm removal,recognizing the role and potential mechanism of ellagic acid in the control of periodontal disease.
