Objective: Based on high-throughput sequencing technology,long non-coding RNA(lnc RNA)expression profiling was performed on prostate cancer(PCa)tissue and corresponding paracancerous tissues,and differentially expressed lnc RNAs in PCa tissues were screened out to explore lnc RNAs that may be associated with PCa.Methods:According to the diagnosis,inclusion and exclusion criteria,3 cases of PCa tissues and corresponding paracancerous tissues were selected and divided into two groups: cancer tissues and paracancerous tissues.The differential expression profiles of lnc RNAs in the two groups were obtained by high-throughput sequencing technology.The informatics method was used to analyze the differences of lnc RNAs in the two groups,and the lnc RNAs with significant differential expression between the two groups were selected,namely DLEU2 and LINC-PINT.Subsequently,the differential expression of the selected lnc RNAs was further verified in the blood samples of 54 cases of PCa and 76 cases of benign prostatic hyperplasia(BPH)patients.At the same time,the correlation between the expression level of lnc RNA and clinicopathological features in PCa patients was explored.Results:(1)Through the analysis of high-throughput sequencing results,it was found that compared with para-cancerous tissues,a total of 2361 lnc RNAs were differentially expressed in PCa tissues,of which 653 were up-regulated and 1708 were down-regulated.(2)The blood sample verification results were consistent with the high-throughput sequencing results,and the expressions of DLEU2 and LINC-PINT were significantly down-regulated in PCa(P(27)0.05).And multivariate Logistic regression showed that the relative expression of DLEU2 in blood(29)1.10 was a protective factor for PCa(P(27)0.05).(3)The decrease of the relative expression of DLEU2 was associated with the occurrence of bone metastasis and the increase of initial PSA level in PCa patients(P(27)0.05).The decrease of the relative expression of LINC-PINT was correlated with higher initial PSA level,Gleason score and worse clinical T stage in PCa patients(P(27)0.05).Conclusion: These prostate cancer-related lnc RNA screened by high throughput sequencing are verified to be consistent with the sequencing results,and are significantly correlated with the clinicopathological features of prostate cancer patients,and may play an important role in the occurrence and development of prostate cancer.
