Objective: To investigate the prognostic value of cell membrane tension-related genes in hepatocellular carcinoma(HCC)and their relationship with immunotherapy.Methods:The relevant data of HCC patients were downloaded from the TCGA database,and cell membrane tension-related genes were obtained by retrieving the literatures.Prognostic risk score model was constructed by using methods such as univariate cox analysis,and the patients were classified into high-risk and low-risk groups based on the risk scores.Kaplan-Meier survival analysis and receiver operating characteristic curve(ROC)were used to verify the results.The nomogram model was constructed by combining risk scores and clinical characteristics,and its performance was evaluated by ROC and other methods.Functional enrichment analysis was performed to find the related pathways and the infiltration level of immune cells.The differences in immune cell infiltration,immune-related pathways,immune checkpoints and immunotherapy between the two groups were also analyzed.Single-cell sequencing data(RNA-seq)of HCC patients were downloaded from the GEO database to analyze the cellular infiltration in HCC tumor microenvironment and the distribution of model genes in each cell type.Results:Constructed a prognostic risk score model consisting of PDCD6 IP,CFL1,and EPO.The model divided all patients into high-risk groups and low-risk groups,suggesting that the overall survival rate was worse in the high-risk group.Risk scores is an independent prognostic factor for HCC patients.The nomogram with risk scores had better predictive performance.Functional enrichment analysis showed that the high-risk group was mainly enriched in pathways related to tumorigenesis and mechanical signal transduction;meanwhile,immune correlation analysis of the two groups indicated that the high-risk group was mainly enriched in pathways related to immune escape;the abundance of immune cell infiltration,expression of immune checkpoints and efficacy of immunotherapy were significantly different between the high-risk groups and low-risk groups(P < 0.05).The sc RNA-seq analysis showed a significant depletion of T cells and NK cells in HCC.Conclusion: In this study,PDCD6 IP,CFL1,and EPO were screened by bioinformatics analysis as potential indicators for prognosis determination of HCC,and the risk scores and nomogram model established based on them are helpful for prognosis assessment and clinical decision making of HCC patients;cell membrane tension-related genes have the value of predicting the efficacy and prognosis of immunotherapy in HCC patients.
