| Objective: 1.Using the latest cohort study of prostate cancer patients,explore the epidemiological trend of prostate cancer;2.The differential expression of non-coding micro RNAs(miRNAs)in prostate cancer tissue and prostatic hyperplasia tissue was detected by miRNA chip technology;3.Study the relationship between the expression of miRNA screened out and prostate cancer;4.To explore the risk factors of prostate cancer;5.To explore the effect of miRNA on the proliferation,invasion,apoptosis and metastasis of prostate cancer cells.Methods: 1.Patients with prostate cancer diagnosed from January1,1975 to December 31,2019 in the SEER database were extracted by SEER stat software for epidemiological trend analysis;2.Carry out miRNA microarray technology and obtain different miRNAs in the tissues of patients with benign prostatic hyperplasia and cancer,and screen the miRNAs to be studied through the microarray results and consulting relevant literature;3.Select the patients with pathological diagnosis in the urology department of the First Affiliated Hospital of Xinjiang Medical University from October 2021 to June 2022,collect their general clinical data,blood samples and prostate tissue samples,test the miRNA expression screened from the patient’s blood samples and prostate tissue samples,and analyze the relationship between prostate cancer and miRNA;4.Analyze the general clinical data and miRNA expression of patients and screen out the risk factors of prostate cancer;5.PC3 prostate cancer cells were transfected with miRNA-222-mimics,and the effects of up-regulation or down-regulation of miRNA expression on the growth,proliferation,invasion,apoptosis and metastasis of prostate cancer cells were detected by CCK8,Transwell,flow cytometry and scratch cell biological experiments.Results: 1.There was no significant change in the age-adjusted incidence of prostate cancer from 1975 to 2019,with an AAPC of 0.45(95% CI:-0.87~1.80).Among the tumor grade,the most significant increase in the incidence of G2 prostate cancer was observed,with an AAPC of 2.99(95% CI:1.47~4.54);the most significant decrease in the incidence of G4 prostate cancer was observed,with an AAPC of-10.39(95%CI:-13.86~-6.77).Among the different tumor stages,the most significant reduction in the incidence of localized prostate cancer was observed with an AAPC of-1.83(95%CI:-2.76~-0.90).Among different races,the incidence of prostate cancer was significantly reduced in American Indian or Alaska Native and Asian or Pacific Islander,with an AAPC of-3.40(95% CI:-3.97~-2.82)and-2.74(95% CI:-4.14~-1.32),respectively.Among the different age groups,the incidence rate was significantly increased in 15-54 and 55-64 age groups with AAPC of 4.03(95% CI:2.73~5.34)and 2.50(95% CI:0.96~4.05),respectively,and significantly decreased in ≥85 age group with AAPC of-2.50(95% CI:-3.43~-1.57);2.The results of miRNA microarray showed that there were many different miRNAs in prostatic hyperplasia and tumor tissue.Four miRNAs,including miRNA-144,miRNA-222,miRNA-1248 and miRNA-3651,were finally selected as research objects through consulting relevant literature;3.This study collected the prostate tissue of 41 patients with benign prostatic hyperplasia and 37 patients with cancer.The results of q RT-PCR showed that the expression of miRNA-222 in prostate cancer tissue was lower than that in non-prostate cancer tissue(P<0.05).The blood samples of 52 patients with prostate cancer and 58 patients with benign prostatic hyperplasia were collected.The results of q RT-PCR showed that the expression of miRNA-222,miRNA-1248 and miRNA-3651 in the blood samples of patients with prostate cancer was lower than that of patients with benign prostatic hyperplasia(P<0.05);4.The analysis results show that the ratio of f/t,the relative expression of miRNA-222 and miRNA-1248 were independent influencing factors of prostate cancer(P<0.05);5.Overexpression of miRNA-222 can significantly change the growth,proliferation,invasion,apoptosis and metastasis of PC3 prostate cancer cells.Conclusion: 1.Although there was no significant change in the overall incidence of prostate cancer in this study,significant changes occurred in the incidence of prostate cancer with different characteristics;2.A batch of new miRNAs related to prostate cancer were found through miRNA microarray results;3.The screened miRNA has a certain relationship with the occurrence of prostate cancer;4.The ratio of f/t,the relative expression of miRNA-222 and miRNA-1248 are important influencing factors of prostate cancer;5.MiRNA-222 can regulate the growth,proliferation,invasion,apoptosis and metastasis of prostate cancer cells. |
