Effect Of Tianxing Jianshenwei Granules On Oxidative Damage Of D-Galactose Induced Brain Tissue In Aging Rats

Objective: In this study,the effects of Tianxing Jianshenwei Granules on oxidative damage of brain tissue and related proteins in D-gal induced aging model rats were observed,and the antioxidant effects and mechanism of Tianxing Jianshenwei granules were discussed,in order to provide scientific basis for the efficacy study of Tianxing Jianshenwei granules.Methods: 60 male SD rats were randomly divided into 6 groups according to body weight,including blank group,model group,positive control group(vitamin E group),Tianxing Jianshenwei granules high-dose group,Tianxing Jianshenwei granules medium-dose group and Tianxing Jianshenwei granules low-dose group,with 10 rats in each group.Except the blank group,the other 5 groups were intraperitoneally injected D-gal solution at the injection dose of 0.5g/kg to establish the aging model.The blank group was intraperitoneally injected 0.9% sodium chloride injection with the same volume ratio,and the modeling time was 42 d.On the15 th day of modeling,administration groups were given intragastric administration,and vitamin E group was given intragastric administration of vitamin E solution at a dose of 27mg/kg.The high,medium and low dose groups of Tianxing Jianshenwei Granules were respectively given 10.8g/kg,5.4g/kg and 2.7g/kg doses of corresponding concentrations of drugs.Blank group and model group were intragastric administration of distilled water with the same volume ratio.The intragastric administration lasted for 28 days.During the experiment,the general state of rats in each group was observed.After the intervention,SOD,GSH-Px and CAT activities and MDA content in brain tissues of rats were detected.The expressions of Klotho protein and nuclear factor E2-related factor 2(Nrf2)were detected by Western Blot.Pathological changes of hippocampus were observed by HE staining.The experimental data were statistically analyzed by SPSS 25.0 software and Excel table.Results:1.General state observation results of rats: compared with the blank group,the rats in model group showed listlessness,inactivity,dull hair,easy to fall off,dry rough skin,lack of elasticity,reduced diet and water intake,slow movement and so on.After drug intervention,the general state of rats in each administration group was improved to varying degrees,and the improvement effect of vitamin E group and Tianxing Jianshenwei granule high-dose group was most obvious.2.Results of SOD activity and MDA content in brain tissue of rats:Compared with blank group,SOD activity in brain tissue of rats in model group was significantly decreased(P<0.01);Compared with model group,SOD activity in brain tissue of rats in vitamin E group and Tianxing Jianshenwei granule groups was significantly increased(P<0.01 or P<0.05).Compared with blank group,MDA content in brain tissue of model group was significantly increased(P<0.01).Compared with model group,MDA content in brain tissue of rats in vitamin E group and Tianxing Jianshenwei granule groups was significantly decreased(P<0.01).3.Results of the activities of GSH-Px and CAT in rat brain tissue:Compared with blank group,the activities of GSH-Px and CAT in brain tissue of model group were significantly decreased(P<0.01);Compared with model group,the activity of GSH-Px in brain tissue of vitamin E group and Tianxing Jianshenwei granule high-dose and medium-dose group was significantly increased(P<0.01 or P<0.05),while the activity of GSH-Px in low-dose group was not significantly changed(P>0.05).The activity of CAT in vitamin E group and Tianxing Jianshenwei granule groups was significantly increased(P<0.01 or P<0.05).4.Expression results of Klotho and Nrf2 protein in brain tissue of rats:Compared with blank group,the expressions of Klotho and Nrf2 in brain tissue of rats in model group were significantly decreased(P<0.01);Compared with model group,the expressions of Klotho and Nrf2 in brain tissue of rats in vitamin E group and Tianxing Jianshenwei granule groups were significantly increased(P<0.01).5.Results of HE staining in hippocampus of rat brain tissue: Compared with blank group,neuron cells in hippocampus of rats in model group were arranged disordered and distributed sparsely,cytoplasmic fusion and swelling,nucleolar shrinkage and axon disappearance,degeneration and necrosis of nerve cells were obvious,some cells became larger and cytoplasmic staining was observed.Compared with model group,pathological damage in hippocampal area of rats in all administration groups was improved to varying degrees,especially in vitamin E group and Tianxing Jianshenwei granule high-dose group.Conclusion:1.Tianxingjianshenwei granules can improve the general state of aging rats,reduce the pathological damage of hippocampus,increase the activities of SOD,GSH-Px and CAT in brain tissue,and reduce the content of MDA.Up-regulated expression levels of Klotho and Nrf2 proteins in brain tissue.2.Tianxingjianshenwei granules may up-regulate the expression levels of Klotho and Nrf2 proteins,activate the antioxidant enzyme system,reduce the accumulation of peroxide,so as to protect brain tissue from oxidative stress damage and delay the aging process.
