ObjectiveIn this experiment,we observe the influence of cross-electro-ape acupuncture(CENA)on cerebral edema,pathological changes of brain tissue,mNSS score and expression of oxidative stress in ICH rats,so as to verify whether the nerves of ICH rats are protected by CENA and its function principle.Method:A total of 128 male SD rats were used as research samples.According to the standard of 32 rats/group,they were divided into four groups: sham acupuncture group,sham operation group,cross elect rophase acupuncture control group and model group,and then each group was divided into four subgroups,namely,1-day group,3-day group,7-day group and 14-day group,which were divided according to different postoperative sampling times.There were 8 rats in each group.ICH model in rats was prepared by two-step injection with auto logous blood.Before operation,the rats abstained from food and water for 8h.ICH Longa score was performed on all rats 2h after operation.The dead rats with scores below the standard line and 1-3 points were included to supplement the modeling of rats.The rats with a score of less than 1 and obvious disturbance of consciousness were excluded,and then successful modeling and models were added to carry out experimental work.Model group:No intervention of any samples,according to the requirements of the establishment of cerebral hemorrhage model.Sham operation group: No blood injection or intervention was performed after anesthesia,fixation,positioning and extraction of animal blood.After the modeling was completed,the same binding treatment was only performed in the electroacupuncture group for 30 min each time.In the sham acupuncture group,the acupuncture point was selected at the original point 2-3cm without meridian or acupoint,and the other methods were the same as those in the acupuncture control group.Acupuncture control group: the method of cross electroacupuncture was used,that is,acupuncture on both sides of "Yifeng" and "Fengchi",and then the positive electrode was connected to "Yifeng",and the negative electrode was connected to the opposite side of "Fengchi".The frequency of electroacupuncture was 2 Hz and the intensity was 0.5 m A.Each treatment lasted 20 min,once a day.Neurological function of rats was evaluated based on the Modified Neurological Severity Score(mNSS)method,and neurological damage was positively correlated with the score.After HE staining,the pathological condition of brain tissue was observed.The water content of rat brain tissue was determined by wet and dry weight method.Stress parameters such as SOD and MDA were measured by ELISA kit.Results:1.mNSS scores were performed at 1 day,3 days,1 week and 2 weeks after the operation:(1)mNSS scores in the sham operation group were much lower than those in the other three groups at each time point(P < 0.01);(2)The neurological defects of sham operation were not obvious at all time points after operation.(3)At 3 days after operation,mNSS scores in sham acupuncture group,model group and cross electric acupuncture group were higher(P < 0.05).(4)mNSS scores in the cross electrical item group were significantly lower than those in the sham acupuncture group and model group at each time point after surgery(P < 0.01).2.At 1,3,1 and 2 weeks after the operation,the water content of the brain tissue of all experimental groups was measured:(1)The water content of the brain tissue of the cross electroacupuncture group was significantly lower than that of the sham acupuncture group and model group at each time point(P < 0.01);(2)In terms of the water content of brain tissue 3 days after operation,the peak value was reached in the cross electroacupuncture group,the sham acupuncture group and the model group(P < 0.05).(3)At each time point,the water content of brain tissue in the sham operation group was significantly lower than that in the other three groups(P < 0.05);(4)The brain edema of the sham operation group was not obvious at each time point after the operation.3.Effects of different intervention methods on pathological morphology of neurons in rats with cerebral hemorrhageHE staining:(1)The shape of brain tissue did not change in the sham operation,the cell shape was normal,the neurons were arranged regularly,there was no necrosis,no cell edema,fragmentation,nuclear shrinkage phenomenon in the nucleus;(2)One and three days after surgery,the pathological injury of brain tissue in the model group was more obvious,including abnormal structure near the focus,serious edema,rapid and massive accumulation of red blood cells,inflammatory cell infiltration,deformation and swelling of nerve cells,and the phenomenon of cell necrosis and apoptosis,and the corresponding time node of the most serious phenomenon was 3 days after surgery.(3)The pathological injury of brain tissue was improved at 1 and 2 weeks after operation,with the improvement of erythrocytolysis,nerve cell edema and inflammatory cell infiltration near the lesion as the direct manifestations.(4)Inflammatory cell infiltration,red blood cell aggregation,and nerve cell swelling were observed in both the sham acupuncture group and the cross electroacupuncture group,with varying degrees.However,the nerve inflammation,cell edema,degeneration and necrosis were reduced in the cross electroacupuncture group compared with the sham acupuncture group and the model group at each time point.4.Quantitative analysis of SOD protein and MDA protein expression in each groupELISA showed that:(1)The expression level of SOD activity was higher in the sham operation group than in the other three groups(P < 0.01).(2)The expression level of lipid peroxides(MDA)in sham operation group was lower than that in the other three groups(P <0.01),but the content of MDA in brain tissue of rats in cross electroacupuncture group was significantly lower than that in sham acupuncture group and model group(P < 0.01).(3)The increase of SOD activity in cross acupuncture group was significantly higher than that in sham acupuncture group and model group(P < 0.01).Conclusion:1.Cross electroacupuncture can reduce pathological injury of brain tissue after cerebral hemorrhage,improve cerebral edema and repair nerve function;2.For rats with intracerebral hemorrhage,cross electroacupuncture can inhibit oxidative stress in brain tissue,which is achieved by stimulating SOD expression and inhibiting MDA expression,so as to avoid the rapid development of brain injury after intracerebral hemorrhage.
