| Objective:We investigated the current situation of anxiety,depression and sleep disorders occurring in brain workers,and further analyzed the effects of anxiety,depression and CLOCK,PER2 and RORA gene polymorphisms and their interactions on sleep disorders,so as to provide scientific reference for reducing the risk of sleep disorders in brain workers.Methods:In this study,2100 brain workers were selected by whole-group sampling at the Health Management Center of Xinjiang Medical University from December 2021 to May2022,and the anxiety,depression,and sleep disorders of the study subjects were evaluated by applying the Self-Assessment Scale for Anxiety(SAS),the Self-Rating Scale for Depression(SDS),and the Pittsburgh Sleep Quality Inventory(PSQI).The CLOCK,PER2,and RORA genes of 748 brain workers(374 randomly selected from each of the sleep disorder and normal sleep groups)were typed by applying the im LDRTM genotyping technique,and the relationship between CLOCK,PER2,and RORA gene polymorphisms and interactions with sleep disorders was analyzed.Results:1)A total of 2100questionnaires were distributed in this study,and 1994 questionnaires were finally determined to be qualified.Among the brain workers,853(42.78%)were males and 1141(57.22%)were females,aged(34.00±8.66)years;2)The anxiety detection rate among brain workers was 31.09%,and the difference in anxiety detection rate among different gender,shift work,education,job title,occupation and monthly income groups was large and statistically significant(P<0.05);among the brain workers(3)The detection rate of sleep disorders among brain workers was 27.88%,among which women,age>45 years,divorced or widowed,shift work,college education or below,junior college education or below,medical workers,and monthly income were all statistically significant(P<0.05).The detection rate of sleep disorders was higher among female,age>45 years,divorced or widowed,shift work,college or below,junior or below,medical personnel,and brain workers with monthly income of 5000-8000 RMB(P<0.05);4)The incidence of sleep disorders differed between groups with different degrees of anxiety and depression(P<0.001);anxiety and depression scores were positively related to PSQI scores(rs=0.626,rs=0.661,P<0.001)and their respective dimensions.scores were positively related to PSQI scores(rs=0.626,rs=0.661,P<0.001);5)The results of multifactorial logistic regression analysis found that age>45 years,shift work,occupation as an executive,monthly income of$5,000-8,000,anxiety and depression all increased the risk of sleep disorders in brain workers(all P<0.05).6)The rs10462028 and rs11932595 loci of the CLOCK gene,the rs934945 loci of the PER2 gene showed significant differences in the distribution of each genotype and allele frequency between the sleep disorder and normal sleep groups(all P<0.05);7)SHEsis software results showed that the A-G and G-A haplotypes of the CLOCK gene differed between the sleep disorder and normal sleep groups(P<0.05);the C-G and T-G haplotypes of PER2 gene were significantly distributed between cases and controls(P<0.05).8)The GMDR software established the best fitted gene-gene interaction model between rs10462028,rs11932595 and rs934945 loci,and the best fitted gene-environment interaction model between rs934945,anxiety and depression;loci The interaction of rs10462028,rs11932595,and rs934945(OR=2.816,95%CI:1.742-4.550)and the interaction of loci rs934945,anxiety,and depression(OR=10.461,95%CI:3.695-29.621)resulted in an increased cerebral sleep disorder higher risk(P<0.05);9)The results of the Bayesian network model of sleep disorders showed a higher degree of influence of anxiety,depression,and shift work on sleep disorders.Conclusion:1)The degree of anxiety and depression was more severe in the brainworkers,and the detection rate of sleep disorders was higher in the anxiety and depression groups.2)The detection rate of sleep disorders in the brainworkers was 27.88%,and age>45 years,shift work,occupation as an executive,monthly income of 5,000-8,000,anxiety and depression could increase the risk of sleep disorders in brainworkers.3)Brainworkers The genotypes and alleles of rs10462028 and rs11932595 loci of CLOCK gene and rs934945 loci of PER2 gene differed among sleep disorder groups;the interaction between loci rs10462028,rs11932595 and rs934945increased the risk of sleep disorders in mental workers population.The interaction between locus rs934945 and anxiety and depression increases the risk of sleep disorders in the cerebral workforce.4)The results of the Bayesian network model of sleep disorders show that anxiety,depression,and shift work have a higher degree of effect on sleep disorders. |
