| Objective: To investigate the expression of lnc RNA KCNQ1OT1 in renal tissues of patients with idiopathic membranous nephropathy(IMN)and its relationship with the number of podocytes and clinical indexes.Methods: 24 patients diagnosed with IMN from February 2022 to June 2022 in the Department of Nephrology,Shengjing Hospital,China Medical University were collected as IMN group,and 22 patients with urological nephrectomy for nephrocalcinoma(renal tissue specimens >5cm from the most distal cut edge of the lesion)were collected as control group.The expression of lnc RNA KCNQ1OT1 was detected by fluorescence in situ hybridization(FISH)and real-time quantitative PCR(q RT-PCR).The podocyte nucleus was labeled with WT1,and the expression level of WT1 was detected by immunohistochemical staining.Meanwhile,clinical data of patients were collected to study the relationship between lnc RNA KCNQ1OT1 expression in the renal tissue of IMN patients and the number of podocytes,serum albumin,24 h urine protein quantification and other clinical indexes.Results: 1.Compared with the control group,there was no significant difference in age,gender,urea,creatinine and estimated glomerular filtration rate(e GFR)between the two groups.The serum albumin level in the IMN group was significantly lower than that in the control group(P<0.05);2.The expression of lnc RNA KCNQ1OT1 was significantly increased in the renal tissue of IMN patients compared with the control group(t=11.911,P<0.05);3.WT1 expression was significantly decreased in the renal tissues of IMN patients compared with the control group(t=-9.777,P<0.05);4.lnc RNA KCNQ1OT1 expression in renal tissue of IMN patients was negatively correlated with the number of podocytes and serum albumin(r=-0.487,P<0.05;r=-0.425,P<0.05),was positively correlated with 24 h urine protein quantification(r=0.482,P<0.05),but was not significantly correlated with creatinine,urea,e GFR and anti phospholipase A2 receptor(PLA2R)antibody levels.Conclusion: 1.The expression of lnc RNA KCNQ1OT1 in renal tissue of IMN patients was higher than that in normal renal tissues.2.The expression of lnc RNA KCNQ1OT1 in renal tissue of IMN patients was negatively correlated with the number of podocytes and serum albumin and positively correlated with 24 h urine protein quantification.3.lnc RNA KCNQ1OT1 may be involved in the development of IMN,providing a new potential target for the treatment of IMN. |
