Relationship Between MicroRNA-29c-3p/DNMT3B/E-cadherin Axis And Clinical Pathological Characteristics Of Gastric Cancer

Objective: To explore the relationship between the expression of mi R-29c-3p,DNMT3 B,E-cadherin and the clinical pathological characteristics of gastric cancer.Methods: The expression and clinical data of mi RNA and m RNA in gastric cancer were downloaded from TCGA database.The expression of mi R-29c-3p,Dnmt3 b and CDH1 as well as the relationship between clinical characteristics and prognosis of gastric cancer were analyzed.The candidate target genes of mi R-29 c were predicted with online databases,including Target Scan,Pictar,mi RTarbase and Starbase,enrichment analysis of gene functions and signal pathway of these candidate target genes were performed with online software DAVID.Gastric cancer tissues and normal tissues were collected to prepare paraffin tissue chip.The expression of mi R-29c-3p in gastric cancer tissues and normal tissues was detected by mi RNAscope.The expression of DNMT3 B and E-cadherin was detected by immunocytochemistry.The relationship between the expression of mi R-29c-3p,DNMT3 B and E-cadherin and the clinical pathological characteristics of gastric cancer was analyzed by chi-square test.The correlation between continuous variables was analyzed by Pearson correlation analysis.The relationship between the expression of mi R-29c-3p,DNMT3 B and E-cadherin and prognosis was analyzed by Log-rank test.Results: TCGA data analysis showed that the expression levels of mi R-29c-3p and CDH1 were significantly decreased(p<0.001)and the expression level of Dnmt3b(p<0.001)was significantly increased in gastric cancer tissue than those in normal tissues;Dnmt3b expression was related to the differentiation degree of gastric cancer,p<0.001;mi R-29c-3p was negatively related to Dnmt3 b expression,mi R-29c-3p was positively related to CDH1 expression,Dnmt3 b was negatively related to E-cadherin expression,p<0.001.Eighteen common candidate target genes of mi R-29 c were predicted with Target Scan,Pictar,mi RTarbase and Starbase databases.GO enrichment analysis showed that mi R-29 c had the activity of DNA methyltransferases,KEGG enrichment analysis showed that the biological function of mi R-29 c was associated with activation of signal pathways of cancer-related mi RNAs.mi RNAscope results showed that the expression level of mi R-29c-3p was decreased in gastric cancer tissues than that in normal tissues,p<0.001,and significantly related to TNM staging(p=0.002)and lymph node metastasis(p=0.0023).Immunohistochemistry results showed that the expression level of DNMT3 B was up-regualted in gastric cancer tissues,p<0.001,and significantly related to the differentiation degree of gastric cancer(p=0.018)and the maximum diameter of the tumor(p=0.03);the expression level of E-cadherin was down-regualted in gastric cancer tissues,p<0.001,and significantly related to the differentiation degree of gastric cancer,p=0.036.Further correlation analysis showed that mi R-29c-3p was negatively related to DNMT3 B expression,p<0.001,and positively related to E-cadherin expression,p<0.001,DNMT3 B was negatively related to E-cadherin expression,p<0.001.Conclusions: 1.mi R-29c-3p,DNMT3 B and E-cadherin were abnormally expressed in gastric cancer tissues,associated with tumor progress,and could be used as tumor markers.2.The expression of mi R-29c-3p,DNMT3 B and E-cadherin of gastric cancer tissues were correlated,and mi R-29c-3p may be involved in the pathogenesis and development of gastric cancer through regulating E-cadherin expression by targeting Dnmt3 b.
