Expression And Significance Of MMP-3 And TIMP-3 In Lens Epithelial Cells In Diabetic Cataract Patients | | Posted on:2024-09-12 | Degree:Master | Type:Thesis | | Country:China | Candidate:X L Zhang | Full Text:PDF | | GTID:2544307094968519 | Subject:Ophthalmology | | Abstract/Summary: | PDF Full Text Request | | ObjectiveThis study was to investigate the expression of matrix metalloproteinase-3(MMP-3)and tissue inhibitor of metalloproteinases-3(TIMP-3)in lens epithelial cells(LECs)in Diabetic cataract patients and Age-related cataract patients,and explore their significance and interrelationship in the development of Diabetic cataract.MethodsA total of 56 diabetic cataract patients in the ophthalmology ward of Jingzhou Central Hospital were selected in the experimental group,and 64 age-related cataract patients in the ophthalmology ward of Jingzhou Central Hospital were selected in the control group.The experimental group and the control group were all subjected to phacoemulsification of cataract combined with intraocular lens implantation by the same experienced surgeon,and the anterior capsule with a diameter of about 5~6mm was torn during the operation.HE and immunohistochemical staining were performed on all anterior capsules to detect the expression of MMP-3 and TIMP-3 proteins in both groups of lens epithelial cells.Results: Each slice was taken at the same microscope magnification,and the obtained images were analyzed in grayscale using Image J.JS software.SPSS21.0 statistical analysis software was used for processing,and the t-test of two independent samples was used in the diabetic cataract group and the age-related cataract group,and the test level was α=0.05.Results1.Compared with age-related cataract group,there were no significant differences in gender,ocular distinction,and intraocular pressure(IOP)in the diabetic cataract group(P>0.05).In terms of age,the age of patients in the diabetic cataract group was significantly lower than that in the age-related cataract group(P<0.01).2.MMP-3 and TIMP-3 are expressed in both diabetic cataract lens epithelial cells and age-related cataract lens epithelial cells.3.The expression of MMP-3 in diabetic cataract lens epithelial cells was significantly higher than that in age-related cataract epithelial cells,and the difference was statistically significant(p<0.05).However,there was no significant difference in the expression of TIMP-3 in diabetic cataract lens epithelial cells and age-related cataract epithelial cells(p>0.05).4.MMP-3 was associated with the expression level of TIMP-3 in the age-related cataract group(r=0.27,p=0.049),while the expression levels of MMP-3 and TIMP-3 in the diabetes-related cataract group were not correlated(r=0.23,p=0.303).5.The ratio of MMP-3/TIMP-3 in LECs between diabetes cataract group and agerelated cataract group was significantly different(P<0.01).Conclusion:1.The expression of MMP-3 in diabetic cataract lens epithelial cells was significantly higher than in age-related cataract lens epithelial cells,indicating that the changes of MMP-3 protein played an important role in the development and progression of cataract in diabetic patients.2.There was no significant difference in the expression of TIMP-3 protein in the two groups of lens epithelial cells,which may prove that TIMP-3 does not directly affect the accelerated change of diabetic cataract progression.3.The imbalance in the expression of MMP-3/TIMP-3 may play an important role in the development and progression of diabetic cataracts. | | Keywords/Search Tags: | Lens Epithelial Cells, Diabetic cataracts, Age-related Cataracts, Matrix Metalloproteinases, Tissue Inhibitor of Metalloproteinases | PDF Full Text Request | Related items |
