Zanthoxylum nitidum(Roxb.)DC,traditionally used as bruises,swollen and painful gums.Benzophenanthridine alkaloids are a major component in Z.nitidum and have been reported to have a wide range of biological activities such as antibacterial,anti-inflammatory,antimalarial,and antitumor.In order to further study the benzophindidine alkaloid components in the Z.nitidum and its inhibitory effect on tumor cells,this paper systematically isolated,identified the alkaloid components in the Z.nitidum and determined its antiproliferative activity against Hep G2 in hepatoma cells.The roots of Z.nitidum were extracted by methanol water reflux,extracted by the forward and reverse silica gel column,medium pressure column combined with bismuth potassium iodide color developer tracking,and 30compounds were systematically separated from the alkaloids in the Z.nitidum.According to the physicochemical properties of the compound,the structure of the compound was identified by NMR,IR,UV,ECD calculations and X-ray single crystal diffraction.Among them,21 benzophindine alkaloids,compounds 1-6 were new compounds,and compounds 16,17,23,24,25,and 27 were separated from Z.nitidum for the first time.The inhibition rate of Hep G2 on hepatoma cells in 21 monomeric compounds was evaluated by CCK-8 method,and the results showed that compounds1,2,4,7,10,12,17 and 20 had certain inhibitory effects on the proliferation of Hep G2in hepatoma cells,and the IC50 values were 10.81,26.93,11.32,43.74,35.24,8.22,41.78 and 22.39,respectively. |
