Metabolomics emphasizes the study of changes in metabolite levels in the whole organism,which is similar to the overall view of traditional Chinese medicine,and provides ideas for explaining the compatibility mechanism of traditional Chinese medicine.In the early stage of the research group,the network pharmacology researched that the active ingredients of Danggui-Yimucao can act on the key targets of improving the complications of type Ⅱ diabetes and analgesic and antiinflammatory.The active ingredients of Buxue Yimu Pill can regulate the core target of hemorrhagic anemia,and it is verified that the compound can significantly downregulate EPO and promote hematopoiesis and blood circulation.On the basis of this study,it is planned to further analyze the molecular mechanism of action of Buxue Yimu Pill and Danggui-Yimucao at the metabolic level by combining metabolomics and network pharmacology.The main contents are as follows:(1)The model of hemorrhagic anemia was constructed in the early stage,and the liver,spleen and kidney of rats were collected,and the metabolites of Buxue Yimu Pill in the treatment of hemorrhagic anemia were identified by nuclear magnetic resonance analysis.3 in liver,9 in spleen and 2 biological markers in kidney were screened out.The metabolic pathways included arginine biosynthesis,Glycine,serine and threonine metabolism,and glycerophospholipid metabolism.Finally,it is shown that the most affected organ of hemorrhagic anemia is the spleen,modern medicine believes that the spleen is the main organ for blood production and blood storage.(2)Through the network pharmacology analysis of the targets of differential metabolites and the active components of Buxue Yimu Pill,48 active components were screened to regulate 23 core targets such as NOS2,ACHE,etc.Integrative analysis of key pharmacological pathways found that 9 of them were consistent,the active ingredient leonurine acts on NOS2 and other targets to regulate arginine biosynthesis;rutin and stigmasterol act on GPX3,ALAS2,etc.,and play an antioxidant role by regulating glutathione metabolism and glycine,serine and threonine metabolism.The role of stress;rutin and other related glycerophospholipid metabolism pathways such as ACHE to play anti-inflammatory effects..(3)Organs such as liver,spleen and uterus in the dysmenorrhea model constructed earlier were collected.Metabolite profiles of organ samples were analyzed by NMR metabolomics.Statistical analysis was used to screen out 10,2,and8 biomarkers in liver,spleen and uterus.Differential metabolites are mainly related to amino acid metabolism,energy metabolism,fatty acid metabolism,tricarboxylic acid cycle and other pathways.Dysmenorrhea is most affected by the liver,and stagnation of liver qi can lead to dysmenorrhea.(4)LC-MS was used to identify 31 chemical components in Danggui-Yimucao,and through network pharmacology analysis of targets related to differential metabolites,it was found that 26 active components such as stigmasterol and leonurine acted on 24 core targets such as NOS3 and GSR.Metabolomics and network pharmacology have a total of 6 pathways.The results showed that the active components of Danggui-Yimucao,such as sitosterol regulate glutathione metabolism and glycine,serine and threonine metabolism to protect cells from oxidative stress.leonurine participates in regulating inflammation.Sexual pathways,regulate inflammation to relieve dysmenorrhea.To sum up,the efficacy of Danggui-Yimucao for dysmenorrhea and Buxue-Yimu Pill for hemorrhagic anemia is significant.The pharmacodynamic material basis of the synergistic effect of Danggui-Yimucao and Buxue Yimu Pill through multicomponent,multi-target and multi-way is explained.This provides a theoretical basis for follow-up research.
