| Objective: 1 To observe the regulatory effect of electroacupuncture and gastronomin on the ability of smell,learning and memory in rats with Alzheimer’s disease(AD).2 To observe the effect of electroacupuncture(EA)on the expression of galylinin receptor 2(Gal R2)and somatostatin(SST)in piriform cortex,olfactory bulb of AD rats.3 To observe the effect of gastrodin(Gas)on the expression of Gal R2 and SST in piriform cortex,olfactory bulb of AD rats.4 To observe the effect of EA combined with Gas on the expression of Gal R2 and SST in piriform cortex,olfactory bulb of AD rats,so as to provid theoretical basis for the clinical treatment of AD.Methods: 40 adult male SD rats,weight of 200~220g,were randomly divided into 5 groups,8/group: normal group(N group),model group(M group),electroacupuncture group(EA group),gastronomin group(Gas group),electroacupuncture combined with gastronomin group(EA+Gas group).N group does not do any processing.M group was dosed with a brain stereotaxic instrument for bilateral microinjection of Aβ1-42,5μl/side in the hippocampus.The EA group was given electroacupuncture "Dazhui","Baihui",and "Zusanli" points,with a frequency of 2Hz,a current intensity of 1m A,and a dense wave for 30 min,qd.The Gas group was injected gastronomin intraperitoneally at a dose of 0.1 ml/100 g,qd,for 4 weeks.The EA+Gas group was given EA and Gas treatment for 4 weeks.The spatial and learning memory abilities of rats in each group were observed and compared by water maze experiments.The changes of olfactory function in each group of rats were analyzed by burying food pellets.Nissl staining and immunohistochemical staining were used to observe the cell morphology and number expression changes of rat piriformis,olfactory bulb Gal R2 and SSTpositive neurons in each group.Western blot was used to detect the expression of piriform,olfactory bulb Gal R2 and SST proteins in each group of rats.Results: 1.The results of water maze detection showed that the average escape latency of rats in M group was significantly prolonged(P<0.05)and the number of platform crossings was significantly reduced(P<0.05).The average evasion latency of rats in each treatment group was shortened(P<0.05)and the number of platform crossings increased(P<0.05),among which the EA+Gas group was the most significant(P<0.05).2.The results of buried food glomerule experiment showed that the time of searching for food globule was significantly prolonged in the M group rats(P<0.05).The search time for food glomerules was shortened in all treatment groups(P<0.05),with EA+Gas being the most significant.3.The results of Nissl staining showed that the number of piriform cortex and olfactory bulb neurons and cell protrusions in the M group were reduced,and the soma was mostly vacuolelike.The number of neurons increased in all treatment groups(P<0.05),and cell morphology improved.Compared with the EA group and the Gas group,the number of piriformis cortex and olfactory bulb neurons in the EA+Gas group increased(P<0.05).4.The immunohistochemical results showed that the number of piriformis cortex and olfactory bulb APP,c-fos and Gal R2-positive cells increased in M group(P<0.05),and the number of SST-positive cells decreased significantly(P<0.05).The number of piriformis cortex and olfactory bulb APP and c-fos-positive cells decreased(P<0.05),and the number of Gal R2 and SST-positive cells increased(P<0.05).Compared with the EA group and Gas group,the number of APP and c-fos positive cells in the EA+Gas group was further reduced(P<0.05),and the number of Gal R2 and SST positive cells was further increased(P<0.05).5.Western blot results showed that the expressions of piriformis cortex and olfactory bulb APP,c-fos and Gal R2 proteins increased(P<0.05)in M group,and the expression of SST proteins decreased significantly(P<0.05).The expression of piriformis cortex and olfactory bulb APP and c-fos proteins decreased(P<0.05),and the expression of Gal R2 and SST proteins increased(P<0.05).Compared with the EA group and the Gas group,the expression of APP and c-fos proteins in the EA+Gas group was further reduced(P<0.05),and Gal R2 and SST proteins increased(P<0.05).Conclusion: 1.Electroacupuncture and gastronomin could improve the sense of smell and learning and memory of AD model rats;2.Electroacupuncture and gastronomin could regulate the expression of piriformis cortex and olfactory bulb-related proteins;3.Electroacupuncture combined with gastronomin had a synergistic effect on the treatment of AD model rats. |
