Objective:Skin cutaneous melanoma(SKCM)is a common skin malignant tumor with high malignancy,easy metastasis and difficulty in early diagnosis.In recent years,the diagnosis and treatment of SKCM have made progress,but the prognosis of patients with advanced SKCM is still poor.This study intends to find biomarkers involved in the development of skin melanoma by bioinformatics,and provide ideas for the diagnosis and treatment of skin melanoma and find the mechanism of its occurrence and development.Methods:Data sets containing gene chip data of human melanoma and normal tissues were retrieved from GEO database,and genes with different expression in melanoma and normal tissue were screened out.The STRING database was used to draw the PPI network of DEGs,and then the Cyto NCA program of Cytoscape software was used to find the core genes.The genes related to the prognosis of melanoma patients in the core genes were screened by GEPIA2 database.Then download the data from TCGA and GTEx to analyze the expression of CA12 in melanoma and normal tissues.The expression of CA12 in skin cutaneous melanoma and surrounding normal skin tissues collected from clinic was detected by q RT-PCR and Western blot.Then,the relationship between the expression of CA12 and the clinicopathological characteristics of patients were analyzed.The biological functions and related pathways of coexpressed genes with CA12 were analyzed by GO and KEGG database.Results:The microarray data of 31 normal tissues and 122 melanoma tissues were obtained from the GEO dataset GSE3189,GSE46517 and GSE15605.After screening the data,136 differentially expressed genes were obtained.By mapping the PPI network,the first 20 genes were selected as core genes.By survival analysis of core genes,seven genes that may affect the overall survival of SKCM patients were obtained.By literature review,we determined to analyze the expression,clinicopathological characteristics and related mechanisms of CA12 in SKCM.The data extracted from TCGA and GTEx databases were analyzed.The results showed that the expression of CA12 in skin melanoma tissue was lower than that in normal tissue(P<0.001);The results of q RT-PCR showed that the expression of CA12 m RNA in SKCM was lower than that in surrounding normal skin tissue;Western blot results showed that the expression of CA12 in protein level in SKCM was lower than that in surrounding normal skin tissue.By drawing ROC curve,we found that the expression of CA12 can be used as a reference factor for diagnosing SKCM.After analyzed the clinical information data,it was found that the expression of CA12 was related to the possible collapse of SKCM(P=0.046).Univariate and multivariate logistic regression showed that T stage,N stage and CA12 expression were independent prognostic factors of SKCM,and CA12 expression was a favorable factor for OS(HR=0.674).GO and KEGG analysis showed that CA12 may regulate the occurrence and development of SKCM by influencing epidermis development,skin development,epidermal cell differentiation,regulating the interaction between cytokine-cytokine receptor interaction,neuroactive ligand-receptor interaction,PI3K-Akt signaling pathway and other ways.Couclusion: The expression of CA12 in SKCM is lower than that in normal tissues.CA12 can be used as a diagnostic marker of SKCM.The expression of CA12 may be related to the rupture of SKCM,and it is an independent prognostic factor of SKCM.CA12 may affect the differentiation and development of epidermal cells by regulating various signaling pathways,then affect the progress of SKCM.
