Objective: Through second-generation sequencing of placental tissues of preeclampsia patients and normal pregnancy patients,differentially expressed CXCL10 was screened out.Real-time quantitative PCR was used to analyze whether there were differences in the expression of CXCL10 in placental tissues of preeclampsia group and normal pregnancy group,so as to clarify the correlation between CXCL10 and preeclampsia.The regulatory ability and mechanism of CXCL10 on trophoblast cells were investigated using HTR-8/Svneo cells.Methods: 1.The placental tissues of 3 normal pregnancies and 3 preeclampsia patients were sequenced and the target genes were screened.2.The placental tissues of 30 normal pregnancies and 30 preeclampsia patients were selected for immunohistochemical test to detect the expression of CXCL10 in placental tissues.3.Placental tissues of 5 normal pregnancies and 5 preeclampsia patients were selected for q PCR assay to determine the m RNA expression level of CXCL10 in placental tissues.4.CXCL10 plasmid and pc DNA no-load plasmid were transfected into HRT-8/Svneo cell lines in vitro(negative control group).5.The effects of CXCL10 overexpression on migration,invasion and proliferation of HRT-8/Svneo cells were detected by cell function assay.6.westernblotting technique was used to detect the protein expression levels of CXCR3,MMP-2 and MMP-9 of CXCL10 receptor after transfection of RT-8/Svneo cells.Results: 1.We obtained 401 differentially expressed genes in the second generation sequencing and selected five differentially expressed genes for validation.After q PCR experiments,CXCL10,the most significantly differentially expressed gene,was selected for further study;2.Immunohistochemistry confirmed the increased expression of CXCL10 in placental tissues of preeclampsia patients compared with that of normal pregnancy;3.PCDNA and PCDNA-CXCL10 were transfected into HTR-8/SVneo cells,and cell scratch assay,CCK8 assay and Transwell invasion assay were performed.The experimental results showed that the proliferation,migration and invasion ability of trophoblast cells were significantly decreased after CXCL10 overexpression compared with negative control group.4.The results of protein imprinting experiment showed that CXCL10 targeting CXCR3 affected the invasion of trophoblast and reduced the expression of MMP-2 and MMP-9.Conclusions: CXCL10 is highly expressed in the placenta of preeclampsia,which has a certain inhibitory effect on the biological behavior of trophoblast cells.CXCL10 can affect the invasion ability of trophoblast cells by inhibiting the expression of MMP-2 and MMP-9,thus participating in the occurrence and development of preeclampsia.
