| Objective: To provide theoretical basis for establishing the quality standard of Anchusa strigosa Banks &Sol,and to ensure the safety of clinical medication.Methods:(1)The whole characters,anatomical characteristics and powder characteristics of Anchusa strigosa Banks & Sol and its adulterants were studied in detail.(2)Taking rutin and caffeic acid as the control,the TLC identification method was established,and the Anchusa strigosa Banks & Sol and its adulterants were compared by TLC.(3)The contents of rutin and caffeic acid in Anchusa strigosa Banks & Sol and its adulterants were determined.(4)Carry out routine item inspection according to relevant contents in Pharmacopoeia,including moisture,ash and acid-insoluble ash,water-soluble extract content and alcoholsoluble extract content.(5)DNA species identification of Anchusa strigosa Banks & Sol and its adulterants,and comparison of ITS2 sequences.Results:(1)The sources of Anchusa strigosa Banks & Sol,Borago officinalis L.and Anchusa strigosa Banks& Sol are different.The leaf base of Anchusa strigosa Banks & Sol grows close to the stem,and the petiole of Borago officinalis L.leaves is obvious.The thickness,length and main vein shape of the three leaves are different.There are differences in the color,shape and size of the three flowers.Dry medicinal materials all have leaves and stems densely pricked by white thorns,which are difficult to distinguish in broken state.The width of root cork layer and phloem and the cross-sectional structure of leaves can be easily distinguished.There is no obvious difference in the identification characteristics between the three powder fibers.(2)Content determination: the rutin content of Anchusa strigosa Banks & Sol is 0.97-1.25 mg/g,and the caffeic acid content is 132-161 μg/g;Rutin content in Borago officinalis L.is 1.12 mg/g,and caffeic acid content is124 μg/g;The rutin content of Anchusa strigosa Banks & Sol is 1.08 mg/g,and the caffeic acid content is136 μg/g.(4)The content of routine project inspection is determined according to the content of China Pharmacopoeia(2020 edition),and the average water content of Anchusa strigosa Banks & Sol is 9.14%-10.83%;The water content of Borago officinalis L.is 8.3%,and that of Anchusa strigosa Banks & Sol is10.79.The average water-soluble extract content of Anchusa strigosa Banks & Sol is 17.46%-26.28%;The water-soluble extract content of Borago officinalis L.is 27.37%;The water-soluble extract of Anchusa strigosa Banks & Sol is 24.39%.The average alcohol-soluble extract content of Anchusa strigosa Banks &Sol ranged from 5.68% to 8.77%.The alcohol-soluble extract content of Borago officinalis L.is 9.23%;The alcohol-soluble extract of Anchusa strigosa Banks & Sol is 8.00%.The total ash content of Anchusa strigosa Banks & Sol ranged from 9.82% to 12.87%,the total ash content of Borago officinalis L.is 9.46%,and the total ash content of Anchusa strigosa Banks & Sol is 12.58%.The acid-insoluble ash content of Anchusa strigosa Banks & Sol is 1.23%-2.51%;Boric acid insoluble ash 1.56%;Oxalic acid insoluble ash content is2.73%.(5)DNA molecular identification showed that the minimum K-2-P distance of Anchusa strigosa Banks & Sol and its adulterants was greater than the maximum distance within the species,and the samples of Anchusa strigosa Banks & Sol in the adjacent method(NJ)tree converged into one vein,and Borago officinalis L.and Anchusa strigosa Banks & Sol showed a single line,which could be distinguished.Conclusion:(1)The basic source and microstructure can be used to distinguish Anchusa strigosa Banks &Sol from its adulterants.(2)The TLC method of Anchusa strigosa Banks & Sol and its adulterants was established and the content was determined.(3)Routine project inspection was carried out,and the range of moisture,ash and extract was obtained.(4)The ITS2 sequence alignment can effectively distinguish and identify Anchusa strigosa Banks & Sol and its adulterants. |
