Preliminary Study On The Quality Standard Of Wubei Gastrointestinal Capsule And Its Anti-Gastric Ulcer Effect

Objective: The quality standard and preliminary stability of Wubei gastrointestinal capsule were investigated,and the HPLC chromatogram was established.The main effective ingredients and key targets of anti-gastric ulcer prescription were studied by network pharmacology.Methods:1.According to the relevant requirements of Chinese Pharmacopoeia on capsules,investigate Wubei gastrointestinal capsules to ensure that routine inspection items are consistent with the requirements of pharmacopoeia.2.According to the customized quality standard of capsules,the preliminary stability of three batches of pilot particle samples was studied;3.The characteristic map of Wubei Gastrointestinal capsule was established by the software "Similarity Evaluation System of Characteristic Map of Traditional Chinese Medicine",and the contents of cinnamaldehyde and dehydrocosinolide were determined at the same time.4.Network pharmacology method was used to collect the active ingredients of galangal,cinnamon and finger citron in prescriptions and their corresponding targets by TCMSP database and Uniprot data,and disease targets related to gastric ulcers were searched by Gene Cards database.Collect intersection targets of drugs and diseases and make Venn diagrams;The protein interaction network and the "TCM--Active ingredient-Target-disease" network were established using STRING database and Cytoscape software.GO and KEGG enrichment analysis of common targets of drugs and diseases was performed using DAVID database.Results: 1.All the checked items are in line with pharmacopoeia provisions;A TLC method was established for the identification of galangal rhizome,finger citron and xylosa sinensis in preparation.2.During the stability test,there was no obvious change in the appearance of Ubei gastrointestinal capsule,and the capsule related inspection items stipulated by pharmacopoeia met the requirements;3.There were 14 common peaks in 10 batches of self-made Wubei gastrointestinal capsule samples,and the similarity was all greater than 0.90.Four compounds were identified,among which peak 1 was cinnamaldehyde,peak 4 was piperine,peak8 was dehydrocostiolactone and peak 9 was costiolactone.4.Using network pharmacology,49 active ingredients and 644 targets were obtained after screening and weight removal;Related target genes obtained from "gastric ulcer" were retrieved.A total of 138 targets were obtained by the intersection analysis of active ingredient targets and disease-related targets.Based on the Degree value,the top 20 targets are selected as key targets.Conclusion: The stability inspection item standard,qualitative identification and content determination method of Wubei gastrointestinal capsule were established,which guaranteed the quality of the preparation and provided an important basis for the production and circulation of the drug in the later period.Finally,network pharmacology was used to predict the active ingredients,target and pathway of the anti-gastric ulcer effect in this prescription.
